摘要
目的优化党参低聚糖脱色纯化工艺,并评价其抗氧化活性。方法在单因素试验基础上,以脱色时间、pH、树脂用量为影响因素,脱色率、回收率的综合评分为评价指标,响应面法优化脱色纯化工艺。再测定低聚糖对DPPH自由基、羟自由基的清除率。结果最佳条件为DM-28型树脂,脱色时间2.3 h,pH4.7,树脂用量3.2 g,脱色率为94.76%,回收率为88.91%,综合评分为92.42分。脱色后,低聚糖含量及对2种自由基的清除率分别为(83.96±2.07)%、(81.27±2.47)%、(17.94±0.30)%。结论脱色可提高党参低聚糖纯度及抗氧化活性。
AIM To optimize the decolorization and purification process for oligosaccharides from Codonopsis Radix and to investigate their anti-oxidant activity.METHODS With decolorization time,pH and resin consumption as influencing factors,comprehensive score for decolorization rate and recovery as an evaluation index,the decolorization and purification process was optimized by response surface method on the basis of single factor test.Subsequently,the scavenging rates of oligosaccharides on DPPH and hydroxyl free radicals were determined.RESULTS The optimal conditions were determined to be DM-28 resin,2.3 h for decolorization time,4.7 for pH,3.2 g for resin consumption,the decolorization rate,recovery and comprehensive score were 94.76%,88.91%and 92.42,respectively.After decolorization,the oligosaccharides’content and scavenging rates on two free radicals were(83.96±2.07)%,(81.27±2.47)%and(17.94±0.30)%,respectively.CONCLUSION Decolorization can enhance the purity and anti-oxidant activity of oligosaccharides from Codonopsis Radix.
