摘要
目的探讨小鼠在急性暴露于捕食者气味——2,4,5-三甲基噻唑(2,4,5-trimethylthiazole,TMT)应激下对丘脑室旁核(paraventricular thalamic nucleus,PVT)-中央杏仁核(central amygdala,CeA)神经通路活动的影响。方法取3只同种系小鼠在CeA注射CTB555,观察丘脑中线核团中投射到CeA的神经元;8只雄性C57BL/6小鼠按照简单随机抽样法分为生理盐水组、TMT(暴露于捕食者气味)组,每组4只,观察急性暴露于TMT应激对小鼠丘脑中线核团及杏仁核的c-Fos表达影响;取6只同种系小鼠在CeA注射RetroBeads,随后按照简单随机抽样法分为生理盐水组、TMT组,每组3只,观察急性暴露于TMT对CeA投射-PVT神经元c-Fos表达影响。取6只同种系小鼠在PVT注射AAV-EF1α-DIO-GCaMp7b、CeA注射AAV/retro-Syn-Cre,待病毒表达3周后,通过向PVT病毒注射位点上方埋置光纤,手术恢复后在体记录小鼠CeA投射-PVT神经元急性暴露于TMT前后的钙信号变化。结果通过向小鼠CeA注射CTB555,发现CeA选择性接受丘脑中线核团大量的PVT投射。与对照组相比,急性暴露于TMT应激下,PVT(P<0.01)及CeA(P<0.05)的c-Fos表达显著增加。小鼠CeA注射RetroBeads后,CeA投射-PVT神经元暴露于TMT后c-Fos表达显著增加(P<0.05)。光纤钙信号记录发现,CeA投射-PVT神经元暴露于TMT后的ΔF/F显著高于暴露于生理盐水的ΔF/F值(P<0.01)。结论CeA投射-PVT神经元在急性暴露于TMT下显著激活。
Objective To investigate the effects of acute exposure to predator odor(2,4,5-trimethylthiazole,TMT)on the activity of the paraventricular thalamic nucleus(PVT)-central amygdala(CeA)neural pathway in mice.Methods CTB555 was injected into CeA in 3 mice of the same lineage,and the neurons projected to CeA in the midline thalamus nuclei were observed.Eight male C57 BL/6 mice were randomly divided into normal saline group and TMT group(exposed to predator odor),with 4 mice in each group.The effects of acute exposure to TMT on c-Fos expression in the midline thalamus nuclei and amygdala of mice were observed.Six mice of the same lineage were injected with RetroBeads in CeA,and then randomly and equally divided into normal saline group and TMT group.The effects of acute exposure to TMT on c-Fos expression in CeA projecting-PVT neurons were observed.The changes in calcium signals of CeA projecting-PVT neurons in vivo under acute exposure to TMT were recorded.AAV-EF1α-DIO-GCaMp7 b was injected in PVT and AAV/retro-Syn-Cre was injected in CeA of 6 mice of the same lineage.In 3 weeks later,optical fiber was embedded above the injection site of PVT to in vivo record the changes in calcium signals of CeA projecting-PVT neurons before and after acute exposure to TMT.Results By injecting CTB555 into CeA,it was found that CeA selectively receives a large number of PVT projections in the midline thalamus nuclei.Compared with the control group,the expression of c-Fos in PVT(P<0.01)and CeA(P<0.05)was significantly increased after acute exposure to TMT.After injection RetroBeads in CeA,c-Fos expression in CeA projecting-PVT neurons was significantly enhanced after exposure to TMT(P<0.05).Fiber photometry revealed that theΔF/F values of CeA projecting-PVT neurons exposed to TMT were significantly higher than those exposed to normal saline(P<0.01).Conclusion CeA projecting-PVT neurons are activated significantly after acute exposure to TMT.
