基于表型性状和SSR分子标记的草地早熟禾遗传多样性分析和分子身份证构建 被引量:12

Genetic Diversity Analysis and Molecular ID Construction of Kentucky Bluegrass Based on Phenotypic Traits and SSR Molecular Markers

摘要 为明确草地早熟禾种质资源的遗传背景,选择26份本土资源和54份国外引进种质进行表型特征和遗传多样性分析,结果表明:叶宽、株高和绿度3个表型性状存在一定变异,但仍有部分种质差异不显著,仅利用表型性状很难进行准确的种质鉴定。采用40对SSR引物序列筛选出多态性扩增效果好的16对引物,对80份草地早熟禾种质资源进行PCR扩增,共扩增出127条带。SSR标记数据显示,16对引物共检测到123个多态性位点,多态性比率96.85%,Nei’s基因多样性指数和Shannon’s多态性信息指数合度的平均值分别为0.34和0.52。遗传相关系数和聚类、主坐标分析结果表明,80份草地早熟禾种质可分为两大类7个亚类。结合表型性状和分子标记结果,构建了80份草地早熟禾种质的字符串、条形码和二维码分子身份证,可为草地早熟禾种质资源的鉴定、合理利用和有效保护提供参考。 In order to clarify the genetic background information of germplasm resource of Kentucky bluegrass(Poa Pratensis L.),26 native and 54 imported germplasm resources were selected for phenotypic characterization and genetic diversity analysis.The results showed that there were some variations in leaf width,plant height and greenness,but there were still some accessions with no significant differences,which indicated that it was difficult to accurately identify accessions by using only phenotypic traits.Based on 40 pairs of SSR primer sequences published by predecessors,16 pairs of primers with good polymorphic amplification effect were screened,and 80 Kentucky bluegrass germplasm resources were amplified by PCR,and a total of 127 bands were amplified.The SSR markers data showed that a total of 123 polymorphic loci were detected by 16 pairs of primers,with a polymorphism rate of 96.85%.The average values of Nei’s gene diversity index(H)and Shannon’s polymorphism information index(I),were 0.34 and 0.52,respectively.NTSYS-PC 2.10e and UPGMA algorithm were used to calculate the genetic correlation coefficients,cluster analysis and principal coordinate analysis.Those 80 accessions were grouped into two major categories,seven subcategories.Based on the results of phenotypic traits and molecular markers,the character string,bar code and two-dimensional code molecular identity cards of 80 Kentucky bluegrass germplasm were constructed,which can provide reference for the identification,rational utilization and effective protection of Kentucky bluegrass germplasm resources.
