摘要
为了提高活疫苗的稳定性,便于活疫苗的保存和运输,本试验将猪乙型脑炎病毒分别与耐热冻干保护剂和传统冻干保护剂进行配比、冻干,制备活疫苗,分别将疫苗置2~8℃保存,定期测定其病毒含量,同时进行25℃、37℃耐老化试验和小鼠免疫原性试验。结果显示,疫苗在2~8℃条件下保存24个月,使用耐热冻干保护剂制备的疫苗病毒含量下降不超过1.0 Log PFU/mL,使用传统冻干保护剂制备的疫苗病毒含量下降超过3.0 Log PFU/mL;25℃常温保存2个月,使用耐热冻干保护剂制备的疫苗病毒含量下降不超过1.0 Log PFU/mL,而使用传统冻干保护剂制备的疫苗病毒含量下降约2.0 Log PFU/mL;37℃保存10 d,使用耐热冻干保护剂制备的疫苗病毒含量下降不超过1.5 Log PFU/mL,使用传统冻干保护剂制备的疫苗下降超过1.5 Log PFU/mL。使用耐热冻干保护剂制备的活疫苗和使用传统冻干保护剂制备的活疫苗对小鼠的免疫原性相当,半数有效剂量分别为10^(-3.12)/0.1 mL和10^(-3.11)/0.1 mL。试验结果表明,耐热冻干保护剂对病毒的保护效力明显优于传统冻干保护剂,具有良好的热稳定性,使用耐热冻干保护剂制备的活疫苗具有良好的免疫原性。
In order to improve the stability of porcine Japanese encephalitis virus live vaccine and facilitate its preservation and transportation,this study prepared live vaccine by lyophilizing and mixing heat-resistant lyophilized protectant and conventional lyophilized protectant,respectively,stored the vaccine at 2-8℃,measured the virus content regularly,tested the aging resistance at 25℃and 37℃,and carried out immunogenicity test in mice.The results showed that when the vaccine was stored at 2-8℃for 24 months,the virus content of the vaccine with heat-resistant lyophilized protectant decreased less than 1.0 Log PFU/mL,while that of the conventional vaccine decreased more than 3.0 Log PFU/mL;when the vaccine was stored at 25℃for 2 months,the virus content of the vaccine with heat-resistant lyophilized protectant decreased less than 1.0 Log PFU/mL,while that of the conventional vaccine decreased about 2.0 Log PFU/mL;when the vaccine was stored at 37℃for 10 days,the virus content of the vaccine with heat-resistant lyophilized protectant decreased no more than 1.5 Log PFU/mL,while that of the conventional vaccine decreased more than 1.5 Log PFU/mL.The immunogenicity of live vaccine prepared with heat-resistant lyophilized protectants was similar to that of live vaccine prepared with conventional lyophilized protectants,with half effective doses of 10^(-3.12)/0.1 mL and 10^(-3.11)/0.1 mL,respectively.Thus,the heat-resistant lyophilized protectant displayed better protective effect on virus than conventional lyophilized protectant,and had good thermal stability;and the preparation of live vaccine with heat-resistant lyophilized protectant maintained good immunogenicity.
