m6A modification of m RNA in skin diseases

摘要 N6-甲基腺嘌呤(N6-methyladenosine,m6A)是真核生物mRNA上最主要的转录后修饰,受甲基转移酶、去甲基酶和m6A结合蛋白调控,对mRNA的剪接、翻译、降解起重要调节作用。皮肤病尤其免疫性皮肤病和皮肤肿瘤发病机制复杂,治疗困难,严重影响患者的生活质量。近年来研究发现m6A及其调控蛋白可影响众多皮肤病的发生和发展。m6A修饰可参与皮肤附件的发育,包括毛囊、汗腺形成。在黑色素瘤、皮肤鳞状细胞癌、梅克尔细胞癌、银屑病等多种皮肤病中,m6A修饰水平显著改变,并影响细胞增殖、分化、迁移等多种生物学过程。m6A及其调控蛋白可能成为皮肤病潜在的分子标志或治疗靶点,在皮肤病的早期诊断、疗效判断、预后预测、基因治疗等方面有广阔的临床应用前景。 N6-methyladenosine(m6A)is the predominant post-transcriptional modification for eukaryotic mRNA.It’s regulated by methyltransferases,demethylases,and m6A binding proteins,and plays an important role in regulating splicing,translation,and degradation of mRNA.Skin diseases,especially immune skin diseases and skin tumors,have a complicated pathogenesis and are refractory to treatment,seriously affecting the patient quality of life.Recent studies have revealed that m6A and its regulatory proteins can affect the development of numerous skin diseases.The m6A modification was found to be involved in skin accessory development,including hair follicle and sweat gland formation.The level of m6A modification was significantly altered in a variety of skin diseases including melanoma,cutaneous squamous cell carcinoma,Merkel cell carcinoma,and psoriasis,and affected a variety of biological processes including cell proliferation and differentiation migration.The m6A and its regulatory proteins may become potential molecular markers or therapeutic targets for skin diseases,and have promising clinical applications in early diagnosis,efficacy determination,prognosis prediction,and gene therapy of skin diseases.
