摘要
为了探究胰岛素样生长因子1(insulin like growth factor 1,IGF1)基因对家禽生长性状的影响,试验利用PCR扩增结合直接测序的方法,对黔东南小香鸡单核苷酸多态性(SNP)进行筛选,并对SNP位点所对应的基因型遗传学进行分析,同时运用SSPS 18.0软件中一般线性模型对基因型与体重、体尺指标进行关联性分析。结果表明:在试验群体中发现IGF1基因第3外显子62 bp处存在1个SNP位点,该位点属于同义突变,并未引起编码氨基酸改变,GA基因型为优势基因型,G等位基因为优势等位基因,该基因座满足Hardy-Weinberg平衡(P>0.05);该SNP位点与黔东南小香鸡体重、体尺性状不存在显著性关联。说明IGF1基因第3外显子62 bp处发生G>A突变,虽然其不影响黔东南小香鸡的生长性状,但可以考虑作为黔东南小香鸡分子遗传标记辅助选育参考位点之一。
In order to explore the effect of insulin-like growth factor 1(IGF1)gene on the growth traits of poultry,the experiment used the method of PCR amplification combined with direct sequencing to screen the single nucleotide polymorphism(SNP)of Qiandongnan Xiaoxiang chickens,and analyzed the genotype genetics corresponding to the SNP locus.At the same time,the GLM model in the SSPS 18.0 software was used to analyze the correlation between genotype and body weight and body size.The results showed that there was a SNP site at 62 bp of exon 3 of IGF1 gene in the experimental population,which was a synonymous mutation and did not cause changes in the encoded amino acid.The GA genotype was the dominant genotype;allele G was the dominant allele;and this locus was in accordance with the Hardy-Weinberg equilibrium(P>0.05).There was no significant association between this SNP locus and the growth traits of Qiandongnan Xiaoxiang chickens.The results suggested that the G/A mutation occured at 62 bp in exon 3 of IGF1 gene.Although it did not affect the growth traits of Qiandongnan Xiaoxiang chickens,it could be considered as one of the reference sites for molecular genetic marker-assisted breeding of Qiandongnan Xiaoxiang chickens.
