谷胱甘肽对杜洛克猪精子冷冻保存效果的影响 被引量:3

Effect of glutathione supplementation on cryopreservation of spermatozoa in Duroc pigs

摘要 为了探讨冷冻保存Ⅰ液中添加不同浓度谷胱甘肽(GSH)对杜洛克公猪精子冷冻保存效果的影响,试验将冷冻-解冻处理的精液分为6组,分别为对照组(冷冻保存Ⅰ液)和试验组(分别在冷冻保存Ⅰ液中添加1.0,1.5,2.0,2.5,3.0 mmol/L的GSH),在液氮中保存,然后检测精子活力、运动参数、质膜完整率、顶体完整率、线粒体活性,活性氧(ROS)水平,丙二醛(MDA)和ATP含量,以及超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)活性,以此来评价解冻后的精液品质。结果表明:与对照组相比,除GSH添加浓度为3.0 mmol/L时精子活力降低外,其他添加浓度精子活力均提高,其中添加浓度为2.5 mmol/L时提高最显著(P<0.05)。与对照组相比,GSH添加浓度为2.5 mmol/L时猪精子在解冻后各项运动参数均显著提高(P<0.05),添加浓度为2.0,3.0 mmol/L时对猪精子部分运动参数也有一定提高,而添加浓度为1.0 mmol/L时各项运动参数降低。与对照组相比,添加不同浓度GSH时精子质膜完整率、顶体完整率、线粒体活性均提高,其中GSH添加浓度为2.5 mmol/L时效果最显著(P<0.05)。与对照组相比,加入不同浓度GSH均可提高精子解冻后SOD、CAT、GSH-Px这三种酶的活性,其中GSH添加浓度为2.5 mmol/L时SOD和GSHPx活性提高最显著(P<0.05),2.0 mmol/L时CAT活性提高最显著(P<0.05)。与对照组相比,GSH添加浓度为2.5 mmol/L时ATP含量提高最显著(P<0.05),ROS水平降低最显著(P<0.05),添加GSH对MDA的抑制效果不明显。说明GSH对猪精子冷冻保存具有一定的改善作用,其中以冷冻保存Ⅰ液中添加2.5 mmol/L GSH的效果最佳,能够显著提高猪精子冷冻保存效果。 In order to explore the effect of different concentrations of glutathione(GSH)added to cryopreservation solutionⅠon the cryopreservation effect of Duroc boar sperm,the frozen thawed semen was divided into 6 groups,namely control group(cryopreservation solutionⅠ)and test groups(cryopreservation solutionⅠsupplemented with 1.0 mmol/L,1.5 mmol/L,2.0 mmol/L,2.5 mmol/L,and 3.0 mmol/L GSH,respectively),which were stored in liquid nitrogen,and then the sperm motility,motility parameters Plasma membrane integrity,acrosome integrity,mitochondrial activity,reactive oxygen species(ROS),malondialdehyde(MDA)and ATP content,as well as superoxide dismutase(SOD),catalase(CAT)and glutathione peroxidase(GSH-Px)activities were used to evaluate the quality of thawed semen.The results showed that compared with the control group,except for the decrease in sperm motility at the GSH addition concentration of 3.0 mmol/L,the sperm motility of the other supplemental concentrations was increased,of which the increase was most significant when the addition concentration was 2.5 mmol/L(P<0.05).Compared with the control group,the motility parameters of boar sperm were significantly improved after thawing at GSH addition concentration of 2.5 mmol/L(P<0.05),and the motility parameters of boar sperm were also improved at 2.0 and 3.0 mmol/L,while the motility parameters of porcine sperm were reduced when the concentration was added to 1.0 mmol/L.Compared with the control group,the plasma membrane integrity,acrosome integrity and mitochondrial activity were improved by GSH with different concentrations,and the effect was most significant when the GSH addition concentration was 2.5 mmol/L(P<0.05).Compared with the control group,the addition of different concentrations of GSH could improve the activities of SOD,was CAT,and GSH-Px after sperm thawing,in which SOD and GSH-Px activities were most significantly increased when GSH addition concentrations of 2.5 mmol/L(P<0.05),and CAT activity was most significantly increased with 2.0 mmol/L of additi
