摘要
从醉鱼草茎部分离获得1株对多种植物病原菌具有良好抑菌活性的内生细菌贝莱斯芽孢杆菌,命名为ZJ1,为进一步评价内生细菌ZJ1的应用潜力,为醉鱼草内生细菌ZJ1的开发利用奠定基础,对其发酵条件和代谢物稳定性进行了研究,并采用比浊法对内生细菌ZJ1的生长曲线进行测定。结果发现,菌株ZJ1的菌体生长量呈先迅速增加后平稳的趋势;采用单因素试验法和正交试验法对其发酵条件进行筛选,再结合方差分析、综合考虑成本,得到菌株ZJ1的最佳发酵条件:YSP培养基为菌株ZJ1的最佳基础培养基,乳糖为最佳碳源,牛肉膏+酵母粉+蛋白胨为最佳氮源,28~30℃为最佳发酵温度,pH值7.0,每500 mL装液量100 mL,接种量1%,转速190 r/min,发酵时间72 h。对菌株ZJ1代谢物进行温度、pH、紫外辐射、光照及室温和冷藏处理下贮存时间抑菌效果及稳定性测定,结果表明,菌株ZJ1发酵液在80℃以上抑菌效果减弱;在pH值1~13范围内仍保有抑菌活性;对紫外光和自然光照射不敏感;在室温和冷藏条件下均可较长时间贮存。
A strain of endophytic bacteria,Bacillus velezensis,with good bacteriostatic activity against a variety of plant pathogens was isolated from the stem of Buddleja lindleyana Fortune,and was named ZJ1.In this study,in order to further evaluate the application potential of endophytic bacteria ZJ1,to lay the foundation for the development and utilization of the endophytic bacteria ZJ1,the fermentation conditions and metabolite stability of ZJ1 were studied.The growth curve of endophytic bacteria ZJ1 was measured by the turbidimetric method.The results showed that the growth of ZJ1 strain showed a trend which was rapidly increasing at first and then stable.The fermentation conditions were screened by single factor test method and orthogonal test method,and then combined with variance analysis and cost,the optimal fermentation conditions of ZJ1 were obtained:the optimal basic culture medium of ZJ1 strain was YSP;the optimal C source was lactose;the optimal N source was beef extract+yeast powder+peptone;the optimal fermentation temperature was 28-30℃;pH was 7.0;the filling volume was 100 mL/500 mL;the inoculation volume was 1%;the rotation speed was 190 r/min;the fermentation time was 72 h.The bacteriostatic effect and stability of ZJ1 strain metabolites were measured under the treatments of temperature,pH,ultraviolet radiation,light,and storage time at room temperature,and refrigeration.The results showed that the bacteriostatic effect of ZJ1 strain fermentation broth was weakened above 80℃;the bacteriostatic activity was retained in the range of pH 1-13;the bacteriostatic compounds were not sensitive to ultraviolet light and natural light;it could be stored for a long time at room temperature and under refrigeration.
