NM23-H2通过下调P-STAT3表达抑制结直肠腺癌的发生及迁移 被引量:3

摘要 目的探讨抑癌基因——NM23-H2对结直肠腺癌迁移能力的影响,以及与Janus激酶-信号转导子与转录激活子(JAK-STAT)信号通路的相关性。方法选取2018年1月至2021年1月湖南省湘潭市第一人民医院收集的病理蜡块80个,其中腺癌35个,癌旁组织20个,淋巴结转移25个。使用免疫组织化学方法进行染色,统计分析3种蜡块P-STAT3、NM23-H2的表达是否具有差异性,将带有NM23-H2基因的重组质粒转染人结直肠腺癌SW620细胞,采用Transwell实验检测NM23-H2对SW620细胞迁移能力的影响,蛋白质印迹法检测稳定转染组和未转染组SW620细胞P-STAT3的表达。结果3种蜡块P-STAT3、NM23-H2表达比较,差异均有统计学意义(P<0.05);NM23-H2能抑制SW620细胞的迁移能力,并能下调SW620细胞P-STAT3的表达。结论NM23-H2抑制结直肠腺癌发生、发展及迁移可能与JAK-STAT信号通路活性下调相关。 Objective To investigate the effect of cancer suppressor gene NM23-H2 on migration ability of colorectal adenocarcinoma and its correlation with Janus kinase-Signal transducer and activator of transcription(JAK-STAT)signaling pathway.Methods A total of 80 pathological wax blocks were collected from the first people′s hospital of Xiangtan city in from January 2018 to January 2021,including 35 cases of adenocarcinoma tissue,20 cases of para-carcinoma tissue,and 25 cases of lymph node metastasis.Immunohistochemical methods was used to staining,and the expressions of P-STAT3 and NM23-H2 in the three wax blocks were statistically analyze.The recombinant plasmid with NM23-H2 gene was transfected into human colorectal adenocarcinoma SW620 cells.Transwell assay was used to detect the effect of NM23-H2 on the migration ability of SW620 cells.Western blotting was used to detect the expression of P-STAT3 in SW620 cells of stable transfection group and non-transfection group.Results There were significant differences in the expressions of P-STAT3 and NM23-H2 among the three wax blocks(P<0.05);NM23-H2 could inhibit the migration ability of SW620 cells and down-regulate the expression of P-STAT3 in SW620 cells.Conclusion The inhibition of NM23-H2 on the occurrence,development and migration of colorectal adenocarcinoma may be related to the down-regulation of JAK-STAT signaling pathway activity.
