摘要
目的:比较吸入氩气(inhaled argon,iAr)和吸入氢气(inhaled hydrogen,iH_(2))对小鼠急性脑缺血再灌注(cerebral ischemia/reperfusion,CIR)损伤后神经功能改善的作用。方法:选取48只健康成年SPF级雄性ICR小鼠,将其随机分为假手术组(Sham组)、局灶性大脑中动脉栓塞(transient middle cerebral artery occlusion,tMCAO)模型组(tMCAO组)、iAr治疗组(tMCAO-iAr组)、iH_(2)治疗组(tMCAO-iH_(2)组)。于再灌注24 h后行神经功能评分,采用2,3,5-氯化三苯基四氮唑(2,3,5-triphenyltetrazolium chloride,TTC)染色观察急性脑梗死损伤程度,同时监测术后体质量和存活率的变化,评估两种气体的神经保护疗效差异。结果:与tMCAO组相比,iAr和iH_(2)治疗均可明显改善神经功能(P<0.01),脑梗死体积、体质量损失均明显减少(均P<0.001),存活率明显提高;组间相比,tMCAO-iAr组在神经功能评分(P<0.01)和皮层梗死体积两个指标上更优于tMCAO-iH_(2)组。结论:再灌注后尽早iAr和iH_(2)对CIR损伤都能起到一定的保护作用,且iAr对神经功能改善和皮层脑区的保护作用更好。
Objective:To compare neuroprotection in experimental mice post-stroke models with inhaled argon(iAr)vs inhaled hydrogen(iH_(2))before cerebral ischemic/reperfusion(CIR)injury.Methods:48 SPF male ICR mice were randomly assigned into sham group,transient middle cerebral artery occlusion(tMCAO)group,tMCAO with iAr(tMCAO-iAr)group,and tMCAO with iH_(2)(tMCAO-iH_(2))group.Using tMCAO mice model,the neurological deficits were compared using neuroscore,cerebral infarct volume in cortex and subcortex by 2,3,5-triphenyltetrazolium chloride(TTC)staining,weight and mortality at 24 h after reperfusion among four groups.Results:Compared with the tMCAO group,both tMCAO-iAr and tMCAO-iH_(2) improved the neuroscore(P<0.01),reduced the infarct volume(P<0.01).Weight loss and mortality were improved in agreement with other neurological functions(P<0.01).For the intergroup comparison,tMCAO-iAr group displayed better outcomes in neuroscores(P<0.01)and smaller infarct area.Conclusion:iAr exerts better neuroprotection than iH_(2) for tMCAO mice.
