摘要
目的:建立超高效液相色谱法(UPLC)测定枸杞中枸杞酸含量的分析方法,并对比不同产地枸杞子中枸杞酸的含量。方法:对不同产地的枸杞样品采用纯水超声提取,提取溶液采用氨基色谱柱ACQUITY UPLC Glycan BEH Amide Column 130 A(2.1×150,1.7μm)检测,以乙腈为流动相A,以0.05 mol/L乙酸铵溶液为流动相B,梯度洗脱,流速0.30 mL/min,柱温25℃,检测波长260 nm。结果:枸杞酸的回归方程为:Y=6.8320X-0.2837(r^(2)=1.0000),枸杞酸在5.6~280.2μg/mL范围内与峰面积积分值呈良好的线性关系;精密度试验测得枸杞酸峰面积RSD为0.18%;重现性试验测得枸杞酸含量平均值为97.5 mg/g,RSD为1.32%;枸杞酸平均加样回收率为94.5%;RSD为1.63%;仪器的精密度好。不同枸杞由于产地和种植不同,枸杞酸含量差异较为明显。结论:该方法简单、灵敏度高、稳定性好、可靠性强,可用于不同枸杞中枸杞酸的含量测定。
Objective To develop a method for determination of 2-O-(β-D-glucopyranosyl)ascorbic acid in Lycium barbarum by ultraperformance liquid chromatography(UPLC),and compare the content of 2-O-(β-D-glucopyranosyl)ascorbic acid in Lycium barbarum from different origins.Method Lycium barbarum samples were extracted by ultrasonic in water.Separations of extraction solution were carried out on an ACQUITY UPLC Glycan BEH Amide Column 130 A(2.1 mm×150 mm,1.7μm).Acetonitrile as mobile phase A,0.05mol/L ammonium acetate solution as mobile phase B,gradient elution,the flow was 0.30 m L/min,the column temperature was 25℃,and the detection wavelength was 260 nm.Result The regression equation of 2-O-(β-D-glucopyranosyl)was Y=6.8320X-0.2837(r^(2)=1.0000),and 2-O-(β-D-glucopyranosyl)ascorbic acid had a good linearity with the peak areas at the range of 5.6-280.2μg/m L.The RSD of peak area of 2-O-(β-D-glucopyranosyl)ascorbic acid was 0.18%by precision test;the mean content of 2-O-(β-D-glucopyranosyl)ascorbic acid was 97.5 mg/g,and the RSD was 1.32%;the average recovery was 94.5%and the RSD was 1.63%.The difference of2-O-(β-D-glucopyranosyl)ascorbic acid content was obvious in Lycium barbarum from different origins and cultivations.Conclusion The method is simple,sensitive,specific,and reproducible,which can be used to determine the content of 2-O-(β-D-glucopyranosyl)ascorbic acid in Lycium barbarum.
