整合素αv通过转化生长因子-β1促进子宫内膜异位基质细胞纤维化的研究 被引量:1

摘要 目的探讨整合素αv对子宫内膜异位基质细胞(ECSC)纤维化的影响,以及其的作用机制。方法采用实时荧光定量聚合酶链反应(qRT-PCR)法检测正常子宫内膜细胞(NESC)和ECSC中整合素αv、转化生长因子-β1(TGF-β1)mRNA的相对表达量;采用Westernblot法检测NESC和ECSC中纤连蛋白(FN)、α平滑肌肌动蛋白(α-SMA)、Ⅰ型胶原蛋白(Col-1)的相对表达量;对ECSC中整合素αv分别进行过表达和低表达、转染整合素αv模拟物和整合素αv小干涉RNA,对ECSC中TGF-β1分别进行低表达、转染TGF-β1小干涉RNA,Westernblot法检测ECSC中FN、α-SMA、Col-1的相对表达量,qRT-PCR法与Westernblot法共同验证整合素αv与TGF-β1的相对表达量关系。结果ECSC中整合素αv与TGF-β1的相对表达量均明显高于NESC(P<0.05);ECSC中FN、α-SMA、Col-1的相对表达量均明显高于NESC(P<0.05);过表达整合素αv可使ECSC的FN、α-SMA、Col-1的相对表达量均显著增加(P<0.05),低表达整合素αv可使ECSC中FN、α-SMA、Col-1的相对表达量均显著降低(P<0.05),整合素αv能够影响TGF-β1的表达(P<0.05);同时过表达整合素αv和低表达TGF-β1可使ECSC中FN、α-SMA、Col-1的相对表达量均显著降低(P<0.05)。结论整合素αv在ECSC中呈高表达水平,过表达后可促进ECSC的纤维化,其作用机制与影响TGF-β1的表达有关。 Objective To investigate the effect of integrinαv on endometriotic stromal cell(ECSC)fibrosis and its mechanism of action.Methods The relative expression levels of integrinαv and transforming growth factor-β1(TGF-β1)mRNA in normal endometrial cells(NESC)and ECSC were detected by real-time quantitative polymerase chain reaction(qRT-PCR).Western blot was used to detect the relative expression levels of fibronectin(FN),α-smooth muscle actin(α-SMA)and typeⅠcollagen(Col-1)in NESC and ECSC.Integrinαv in ECSC was overexpressed and underexpressed,respectively;transfected with integrinαv mimics and integrinαv small interfering RNAs,respectively;down-expressed TGF-β1 in ECSCs,transfected TGF-β1 small interfering RNAs and detected the relative expression of FN,α-SMA,Col-1.Western blot was used to detect the relative expression levels of FN,α-SMA and COL-1 in ECSC.The relative expression levels of integrinαv and TGF-β1 were verify by the qRT-PCR and Western blot.Results The relative expressions of integrinαv and TGF-β1 in ECSC were significantly higher than those in NESC(P<0.05).The relative expressions of FN,α-SMA and Col-1 in ECSC were significantly higher than those in NESC(P<0.05).Overexpression of integrinαv can significantly increase the relative expressions of FN,α-SMA and Col-1 in ECSC(P<0.05).Underexpression of integrinαv can significantly decreased the relative expression levels of FN,α-SMA,Col-1(P<0.05),and integrinαv could affect the expression of TGF-β1(P<0.05).When overexpression of integrinαv and underexpression of TGF-β1 at the same time could significantly decreased the expression of FN,α-SMA and Col-1 in ECSC(P<0.05).Conclusions Integrinαv is highly expressed in ECSC.Its overexpression can promote the fibrosis of ECSC and its mechanism is related to the expression of TGF-β1.
