摘要
以葛根新品种‘桂粉葛1号’组培苗为试验材料,利用正交实验设计L(3~4)分析6-BA、NAA、Me JA和蔗糖浓度对葛根试管块根诱导的影响,筛选适宜的葛根试管块根诱导培养基配方;利用组织解剖学研究葛根试管块根不同形成时期的形态结构及淀粉粒发育。结果表明:葛根试管块根诱导的较优配方为MS+2 mg·L^(-1)6-BA+0.5 mg·L^(-1)NAA+2.2 mg·L^(-1)Me JA+80 g·L^(-1)蔗糖+6.0 g·L^(-1)琼脂(p H 5.8),在该配方诱导条件下,试管块根的诱导率达95%以上。试管块根的膨大增粗是由于维管形成层产生大量的次生木质部薄壁细胞以及这些细胞生长的结果。随着诱导时间的推移,试管块根淀粉粒不断积累,后期形成了大量的淀粉粒,且外层薄璧细胞中的淀粉体发生早、数量多、体积较大。
To obtain suitable induction of tuberous root in vitro culture of kudzu (Pueraria thomsonii‘Guifenge No.1’),nodal explants were used to study the effect of 6-BA,NAA,Me JA,and sucrose with an orthogonal experimental design.We studied the morphological structure of tuberous root in vitro culture of kudzu at different stages through morphological anatomy.The results suggested that the optimal conditions for inducing in vitro tuberous root cultures was MS basal medium containing 2.0 mg·L^(-1)6-BA,0.5mg·L^(-1)NAA,2.2 mg·L^(-1)Me JA,80.0 g·L^(-1)sucrose and 6.0 g·L^(-1)agar (p H 5.8),with a tuberization rate of more than 95%.The expansion and thickening of tubers were due to the production of a large number of secondary xylem parenchyma cells in the vascular cambium and the growth of these cells.With the passage of induction time,the starch granules of in vitro tuberous root accumulated continuously,a large number of starch granules were formed in the later stage,and the amyloplasts in the outer thin wall cells occurred early,large in number and large in volume.
