长链非编码RNA lncRNA MEG8通过靶向miR-181a-5p调控脑胶质瘤细胞的增殖与转移潜能 被引量:2

Long non-coding RNA lncRNA MEG8 regulates proliferation and metastatic potential of cerebral glioma cells by targeting miR-181a-5p

摘要 目的阐明lncRNA MEG8在脑胶质瘤发生中的潜在作用和调控机制。方法公共数据库分析lncRNA MEG8在脑胶质瘤的表达量及与肿瘤预后关系, 荧光定量PCR验证lncRNA MEG8和微RNA(miRNA, miR)-181a-5p在本中心脑胶质瘤生物样本库中的表达水平。质粒和miRNA模拟物分别被转染进入脑胶质瘤细胞上调lncRNA MEG8和miR-181a-5p表达, 通过CCK-8增殖、Transwell实验、双荧光素酶报告检测lncRNA MEG8和miR-181a-5p对脑胶质瘤细胞增殖及转移能力的影响及潜在的调控机制。结果与邻近正常组织相比, lncRNA MEG8在脑胶质瘤组织中呈低表达, 其表达水平与患者的无瘤生存期密切相关。上调lncRNA MEG8的表达可显着抑制脑胶质瘤细胞的增殖能力、降低转移能力;miR-181a-5p在脑胶质瘤组织中呈高表达, 其表达水平与lncRNA MEG8呈负相关关系, 且miR-181a-5p可逆转lncRNA MEG8的抑癌功能。结论 LncRNA MEG8通过表达下调miR-181a-5p的表达水平从而抑制脑胶质瘤细胞增殖及转移能力, 是脑胶质瘤的潜在治疗靶点。 Objective To elucidate the potential action and regulatory role of lncRNA MEG8 in development of cerebral glioma.Methods Public databases were used to analyze the expression of lncRNA MEG8 in cerebral glioma and its relationship with tumor prognosis.Fluorescence quantitative PCR was used to verify the expression levels of lncRNA MEG8 and miR-181a-5p in the cerebral glioma biobank at our center.Plasmids and microRNA(miRNA)mimics were transfected into the glioma cells to up-regulate the expression of lncRNA MEG8 and miR-181a-5p,respectively.The effects of lncRNA MEG8 and miR-181a-5p on the proliferation and metastasis of glioma cells and their potential regulatory roles were evaluated by CCK-8,Transwell,and dual luciferase reporter assays.Results Compared with normal adjacent tissues,lncRNA MEG8 was lowly expressed in glioma tissues,and the expression level was closely associated with tumor-free survival in the patients.Up-regulating the expression of lncRNA MEG8 could significantly inhibit the proliferation and reduce the metastasis of the cerebral glioma cells.miR-181a-5p was highly expressed in glioma tissues,and the expression level was negatively correlated with lncRNA MEG8 expression.miR-181a-5p could reverse the tumor suppressor function of lncRNA MEG8.Conclusion LncRNA MEG8 can inhibit the proliferation and metastasis of cerebral glioma cells by down-regulating the expression level of miR-181a-5p,and therefore appears to be a potential therapeutic target for cerebral glioma.
