损伤-修复模型在药物作用癌细胞上的应用

摘要 在进化模型Crow-Kimura模型的基础上,引入药物剂量和修复作用,建立了药物作用于癌细胞的损伤-修复模型,研究了在几种药物作用下的不同癌细胞各状态的相对浓度随药物剂量的变化。模型预测了MCF-7细胞对核桃蛋白胃酶分解产物、bel 7402细胞对β-胡萝卜素氧化降解产物A以及Hela细胞、U20S细胞和SMMC-7721细胞对藏药纤毛婆婆钠乙酸乙酯提取物的半抑制浓度,给出了相应模型参数的取值,理论结果与实验数据高度符合。模型对于“肩型”和“马鞍型”相对存活率-剂量曲线关系的出现给予了合理解释,为之后药物作用于癌细胞的理论研究提供了新的思路,为相关药物作用于癌细胞的实验提供了参考。 On the basis of the Crow-Kimura model of the evolution model,the drug dosage and the repair effect are intro⁃duced to establish the damage-repair model of the drugs on the cancer cells,and the relative concentrations of different cancer cells under the action of several drugs is studied with the drug dosage.The model predicts IC50 of the MCF-7 cells treated with pepsin hydrolysate of walnut protein,bel 7402 cells treated withβ-carotene oxidation products A,and Hela cells,U20S cells and SMMC-7721 cells treated with ethyl acetate extract from Veronica ciliata Fisch.And then the val⁃ues of the corresponding model parameters are given.The theoretical results are highly consistent with the experimental data.The model provides a reasonable explanation for the emergence of the relative survival-dose curve relationship of“shoulder type”and“saddle type”.It provides a reference for the theoretical study of the drugs action on cancer cells and provides guidance for the experiment of the related drugs action on cancer cells.
