摘要
目的探讨仙茅苷调控circ_0004104对氧化低密度脂蛋白(ox-LDL)诱导的血管内皮细胞(HUVEC)损伤的影响。方法将人脐静脉血管内皮细胞(HUVEC)分为对照(con)组、ox-LDL组、ox-LDL+仙茅苷(0.5、5、50μmol/L)组、ox-LDL+si-NC组、ox-LDL+si-circ_0004104组、ox-LDL+仙茅苷50μmol/L+pcDNA-circ_0004104组。流式细胞术检测HUVEC凋亡率。试剂盒分析HUVEC内超氧化物歧化酶(SOD)活性、丙二醛(MDA)水平以及乳酸脱氢酶(LDH)释放量。RT-qPCR检测circ_0004104表达。结果与con组比较,ox-LDL组HUVEC凋亡率、MDA水平、LDH释放量、circ_0004104相对水平显著升高(P<0.05),SOD活性显著降低(P<0.05)。与ox-LDL组比较,ox-LDL+仙茅苷(0.5、5、50μmol/L)组HUVEC凋亡率、MDA水平、LDH释放量、circ_0004104相对水平显著降低(P<0.05),SOD活性显著升高(P<0.05)。与ox-LDL+si-NC组比较,ox-LDL+si-circ_0004104组HUVEC凋亡率、MDA水平、LDH释放量显著降低(P<0.05),SOD活性显著升高(P<0.05)。与ox-LDL+仙茅苷50μmol/L组比较,ox-LDL+仙茅苷50μmol/L+pcDNA-circ_0004104组HUVEC凋亡率、MDA水平、LDH释放量显著升高(P<0.05),SOD活性显著降低(P<0.05)。结论仙茅苷通过下调circ_0004104表达可减轻ox-LDL诱导的HUVEC凋亡和氧化应激损伤。
Objective To investigate the effects of curculigoside regulating circ_0004104 on ox-LDL induced vascular endothelial cells(HUVEC)injury in vitro.Methods The HUVECs were divided into control group,ox-LDL group,ox-LDL+curculigoside(0.5,5,50μmol/L)groups,ox-LDL+si-NC group,ox-LDL+si-circ_0004104 group,ox-LDL+curculigoside 50μmol/L+pcDNA-circ_0004104 group.Flow cytometry was used to detect cell apoptosis rate;the kits detected the activity of superoxide dismutase(SOD),and the levels of malondialdehyde(MDA)and the release of lactate dehydrogenase(LDH)in HUVECs;RT-qPCR detected the expression of circ_0004104.Results Compared with those in control group,the apoptosis rate,and the levels of MDA,LDH and circ_0004104 in ox-LDL group were significantly increased(P<0.05),however,SOD activity was significantly decreased(P<0.05).Compared with those in ox-LDL group,the apoptosis rate,and the levels of MDA,LDH and circ_0004104 in ox-LDL+curculigoside(0.5,5,50μmol/L)groups were significantly decreased(P<0.05),but,the SOD activity was significantly increased(P<0.05).Compared with those in ox-LDL+si-NC group,the apoptosis rate,and the levelsof MDA,LDH in ox-LDL+si-circ_0004104 group were significantly decreased(P<0.05),however,SOD activity was significantly increased(P<0.05).Compared with those in ox-LDL+curculigoside 50μmol/L group,the apoptosis rate,and the levels of MDA,LDH in ox-LDL+curculigoside 50μmol/L+pcDNA-circ_0004104 group were significantly increased(P<0.05),but,SOD activity was significantly decreased(P<0.05).Conclusion Curculigoside can relieve the ox-LDL induced HUVEC apoptosis and oxidative stress injury by down-regulating the espression of circ_0004104.
