摘要
编码含MATH结构域(Meprin and TRAF homology domain)的基因目前在油菜中研究较少。本研究以甘蓝型油菜‘湘油15’(XY15)种子发育35 d抑制差减(suppression subtractive hybridization,SSH)文库为基础,进一步筛选获得实际编码中有两个连续MATH结构域的功能未知基因,该基因全长DNA序列为1800 bp,我们将其命名为BnaM154。将目的片段克隆后插入表达载体pFGC5941-Nap中,成功构建了pFGC5941-Nap::BnaM154过表达载体。利用农杆菌介导的甘蓝型油菜下胚轴遗传转化方法,成功获得8株稳定遗传的转基因苗,为进一步研究BnaM154基因在甘蓝型油菜中的功能提供了科学依据。
Little is known about the MATH(Meprin and TRAF homology)domain encoding genes in Brassica napus.Based on the suppression subtractive library(SSH)of Brassica napus’Xiangyou 15’(XY15)seeds at different development stages,a specific gene fragment encoding two MATH domains with a length of 1800 bp was scre-ened and then named BnaM154.After cloning,the target fragment was forward inserted into the overexpression vector pFGC5941-Nap,and the overexpression vector pFGC5941-Nap::BnaM154 was successfully constructed.Using Agrobacterium-mediated genetic transformation of hypocotyls in Brassica napus,eight stable transgenic plants were finally obtained,providing a good and stable material basis for further study on the function of BnaM154in Brassica napus.
