摘要
目的探讨葛根异黄酮对D-半乳糖致亚急性衰老小鼠T细胞功能及端粒酶活性的影响。方法将BALB/c雄性小鼠随机分为4组,即对照组、模型组及葛根异黄酮低、高剂量组,每组15只。模型组及葛根异黄酮低、高剂量组小鼠每日背部皮下注射D-半乳糖溶液(160 mg·kg^(-1));造模同时,葛根异黄酮低、高剂量组小鼠灌胃给予50、150 mg·kg^(-1)葛根异黄酮,每天1次,连续给药3个月。分离培养小鼠脾脏T细胞;采用CCK-8法检测细胞增殖;ELISA法检测细胞培养上清中白细胞介素2(IL-2)的含量;流式细胞术检测细胞凋亡率;端粒重复片断扩增(TRAP)-ELISA法检测T细胞端粒酶的相对活性(RTA)。结果与对照组比较,模型组T细胞的增殖率、IL-2分泌水平及活化T细胞的RTA明显降低(P<0.05),细胞凋亡率明显升高(P<0.05)。与模型组比较,葛根异黄酮高剂量组T细胞的增殖率、IL-2分泌水平及活化T细胞的RTA明显升高(P<0.05),细胞凋亡率明显降低(P<0.05)。结论葛根异黄酮能增强衰老小鼠T细胞的活化增殖,促进IL-2分泌,降低T细胞凋亡率并提高端粒酶活性。
Objective To investigate the effect of pueraria isoflavone on T cell function and telomerase activity in Dgalactose induced subacute senile mice.Methods BALB/c male mice were randomly divided into 4 groups,which are the control group,the model group and the pueraria isoflavone low-and high-dose groups,with 15 mice in each group.The mice in the model group and pueraria isoflavone low-and high-dose groups were injected with Dgalactose solution(160 mg·kg^(-1))subcutaneously on the back daily;at the same time,the mice in the pueraria isoflavone low-and high-dose groups were given 50 and 150 mg·kg^(-1)of pueraria isoflavone by gavage once a day for consecutive 3 months.T cells from mouse spleen were isolated and cultured;CCK-8 was used to detect the cell proliferation;ELISA was used to detect the interleukin 2(IL-2)in cell culture supernatant;flow cytometry was used to detect the cytometryapoptosis;telomeric repeat amplification protocol(TRAP)-ELISA was used to detect the relative telomerase activity(RTA)of T cells.Results Compared with the control group,the proliferation rate,IL-2 secretion and RTA of T cells in the model group were significantly decreased(P<0.05),and the apoptosis rate was significantly increased(P<0.05).Compared with the model group,the proliferation rate,IL-2 secretion and RTA of T cells in the pueraria isoflavone high-dose group were significantly increased(P<0.05),and the apoptosis rate was significantly decreased(P<0.05).Conclusions Pueraria isoflavones has effect in enhancing the activation and proliferation of T cells in senile mice,promoting IL-2 secretion,reducing apoptosis and increasing telomerase activity.
