摘要
目的采用基于超高效液相色谱-四极杆-飞行时间质谱联用(UPLC-QTOF-MS)技术的代谢组学方法探讨与胃热证相关的潜在生物标志物,以及左金丸干预大鼠胃热证的作用机制。方法通过灌胃辣椒水煎液及无水乙醇复制大鼠胃热证模型;灌胃左金丸水提液(3.5 g·kg^(-1))进行干预,每天1次,连续4 d。采用UPLCQTOF-MS技术建立大鼠血清代谢指纹图谱;通过主成分分析(PCA)、正交偏最小二乘判别分析(OPLS-DA)筛选左金丸干预胃热证大鼠的血清潜在生物标志物,并进行代谢通路分析。结果正、负离子模式下分别筛选出与胃热证相关的27、14个潜在生物标志物。左金丸能够有效逆转胃热证大鼠的血清代谢紊乱,并对11个(正离子模式)、7个(负离子模式)与胃热证相关的潜在生物标志物产生显著回调作用,分析得到4条密切相关的代谢通路。结论左金丸可能通过调节维生素B6代谢、烟酸和烟酰胺代谢、甘油磷脂代谢、鞘氨醇代谢通路,减轻炎性反应,促进胃黏膜修复,从而发挥对胃热证模型大鼠的保护作用。
Objective To investigate the potential biomarkers associated with stomach heat syndrome and the mechanism of Zuojin Pills in intervening with stomach heat syndrome in rats via metabonomics methodology based on ultra performance liquid chromatography-quadrupole-time-of-flight mass spectrometry(UPLC-QTOF-MS).Methods The rat model of stomach heat syndrome was replicated by gavage of chili pepper decoction and absolute ethyl alcohol;the intervention was carried out by gavage of Zuojin Pills extract(3.5 g·kg^(-1))once a day for consecutive 4 days.The serum metabolic fingerprints of rats were established by UPLC-QTOF-MS technique;principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA)were used to screen potential serum biomarkers of Zuojin Pills in rats with stomach heat syndrome,and metabolic pathway analysis was performed.Results A total of 27 and 14 potential biomarkers associated with stomach heat syndrome were identified in positive and negative ion modes,respectively.Zuojin Pills has effect in reversing serum metabolic disorders in rats of stomach heat syndrome,with significant effect in modulating 11(positive ion mode)and 7(negative ion mode)potential biomarkers associated with stomach heat syndrome,and four closely related metabolic pathways were analyzed.Conclusion Zuojin Pills may exert protective effects on rats in a model of stomach heat syndrome by regulating vitamin B6 metabolism,niacin and nicotinamide metabolism,glycerophospholipid metabolism and sphingomyelin metabolism pathways,reducing inflammatory responses and promoting gastric mucosal repair.
