摘要
目的:研究多西他赛联合四君子汤对RM-1前列腺癌荷瘤小鼠的肿瘤抑制作用及免疫功能的影响。方法:C57BL/6雄性小鼠按随机数字表法分为空白对照组、荷瘤模型组、多西他赛组、四君子汤组、多西他赛联合四君子汤组。除空白对照组外,其余各组建立RM-1前列腺癌荷瘤小鼠模型。各给药组小鼠给予相应干预,空白对照组和荷瘤模型组小鼠同时给予等量的生理盐水灌胃、等量的生理盐水腹腔注射,1次/d,各组小鼠均连续干预10 d。通过瘤体体积、瘤体质量比较抑瘤作用。流式细胞仪检测淋巴结树突状细胞(DCs)细胞标志物(CD11c、CD80、CD86)、肿瘤瘤体T淋巴细胞标志物(CD3、CD4、CD8)、脾脏记忆T淋巴细胞标志物(CD3、CD8、CD44、CD62L)的表达情况。ELISA法检测血清CD4^(+)T、CD8^(+)T淋巴细胞表达情况。结果:与荷瘤模型组比较,多西他赛组、四君子汤组及多西他赛联合四君子汤组小鼠瘤体体积、瘤体质量均明显减小(P<0.05),其中多西他赛联合四君子汤组抑瘤率最高(86.65%)。与荷瘤模型组比较,多西他赛联合四君子汤组小鼠淋巴结CD11c+CD80+细胞比例、CD11c+CD86+细胞比例、CD11c^(+)CD80^(+)CD86^(+)T细胞比例明显升高(P<0.05);瘤体CD3^(+)CD8^(+)T细胞比例明显升高(P<0.05),CD3^(+)CD4^(+)T细胞比例明显降低(P<0.05);脾脏CD3^(+)CD8^(+)CD44^(-)CD62L^(+)T细胞比例明显升高(P<0.05),脾脏CD3^(+)CD8^(+)CD44^(+)CD62L^(-)T细胞比例明显降低(P<0.05);血清CD4^(+)T细胞比例明显升高(P<0.05),血清CD8^(+)T细胞比例明显降低(P<0.05)。结论:多西他赛联合四君子汤对RM-1前列腺癌荷瘤小鼠具有协同抑瘤作用,其通过增强化疗荷瘤小鼠树突状细胞提呈作用、激活T淋巴细胞,调动机体固有免疫细胞、适应性免疫细胞,提高化疗荷瘤小鼠的免疫功能。
Objective:To study the effect of docetaxel combined with Sijunzi decoction on tumor inhibition and immune function in RM-1 prostate cancer-bearing mice.Methods:C57BL/6 male mice were randomly divided into blank control group,tumor-bearing model group,docetaxel group,Sijunzi decoction group,and docetaxel combined with Sijunzi decoction group.Except for the blank control group,the other groups established the RM-1 prostate cancer tumor-bearing mouse model.The mice in each administration group were given corresponding intervention.The mice in the blank control group and the tumor-bearing model group were given the same amount of normal saline by gavage and the same amount of normal saline by intraperitoneal injection,once daily.The mice in each group were continuously intervened for 10 days.The tumor inhibitory effect was compared by tumor volume and tumor mass.The expression of DCs markers(CD11c,CD80,CD86),tumor T lymphocyte markers(CD3,CD4,CD8)and spleen memory T lymphocyte markers(CD3,CD8,CD44,CD62L)were detected by flow cytometry.The expression of serum CD4^(+)T and CD8^(+)T lymphocytes was detected by ELISA.Results:Compared with the tumor-bearing model group,the tumor volume and tumor weight of mice in docetaxel group,Sijunzi decoction group and docetaxel combined with Sijunzi decoction group were significantly decreased(P<0.05),and the tumor inhibition rate of docetaxel combined with Sijunzi decoction group was the highest(86.65%).Compared with the tumor-bearing model group,the proportions of CD11c+CD80+cells,CD11c+CD86+cells and CD11c^(+)CD80^(+)CD86^(+)T cells in the lymph nodes of mice in the docetaxel combined with Sijunzi decoction group were significantly increased(P<0.05).The proportion of CD3^(+)CD8^(+)T cells was significantly increased(P<0.05),and the proportion of CD3^(+)CD4^(+)T cells was significantly decreased(P<0.05).The proportion of CD3^(+)CD8^(+)CD44^(-)CD62L^(+)T cells in spleen was significantly increased(P<0.05),and the proportion of CD3^(+)CD8^(+)CD44^(+)CD62L^(-)T cells in spleen was sig
