The expression and clinical significance of TMED5 and miR-652-3P in primary liver cancer

摘要 目的:分析原发性肝癌(PLC)组织中微小RNA-652-3P(miR-652-3P)和跨膜p24运输蛋白5(TMED5)的表达水平,并探讨其与患者临床病理特征及预后的关系。方法:选取2015年12月至2018年12月行手术治疗的PLC患者82例为研究对象,取手术切除的肝癌组织为观察组,同时选取82例患者手术切除的边缘正常组织(距离癌组织>5 cm)为对照组。采用实时荧光定量PCR法检测所有组织样本miR-652-3P和TMED5 mRNA表达水平,采用Pearson法分析miR-652-3P与TMED5的相关性。分析miR-652-3P和TMED5与患者临床病理特征的关系,Kaplan-Meier分析miR-652-3P和TMED5表达情况与PLC患者生存情况的关系,采用Cox法分析影响PLC患者预后的因素。结果:与对照组组织相比,观察组组织TMED5 mRNA相对表达水平较高(P<0.05),miR-652-3P表达水平较低(P<0.05);PLC组织中miR-652-3P与TMED5呈负相关(P<0.05);miR-652-3P和TMED5与TNM分期、淋巴结转移、淋巴血管间隙浸润相关(P<0.05),与患者年龄、性别、肿瘤大小及分化程度无关(P>0.05);Kaplan-Meier法显示miR-652-3P高表达者平均生存时间显著长于miR-652-3P低表达者,TMED5低表达者平均生存时间显著长于TMED5高表达者;多因素分析表明,淋巴结有转移、miR-652-3P低表达和TMED5高表达是影响PLC患者不良预后的独立危险因素(P<0.05)。结论:miR-652-3P在PLC组织中表达显著下降,TMED5在PLC组织中表达显著上升,二者水平与淋巴结转移、TNM分期、淋巴血管间隙浸润显著相关,可能是PLC患者预后评估有价值的生物指标。 Objective:To analyze the expression levels of microRNA-652-3P(miR-652-3P)and transmembrane transport protein 5(TMED5)in primary liver cancer tissues,and to explore their relationship with the clinicopathological characteristics and prognosis of patients relationship.Methods:Eighty-two patients with primary liver cancer who underwent surgical treatment in our hospital from December 2015 to December 2018 were selected as the research objects,the surgically removed liver cancer tissue was selected as the experimental group,and the tissue(>5 cm from cancer tissue)from 82 patients with normal surgical margins was used as the control group.Real-time fluorescent quantitative PCR method was used to detect the expression levels of miR-652-3P and TMED5 mRNA in all tissue samples,and Pearson method was used to analyze the correlation between miR-652-3P and TMED5.The relationships between miR-652-3P and TMED5 and the clinical pathological characteristics of patients were analyzed,Kaplan-Meier was used to analyze the relationship between the expression of miR-652-3P and TMED5 and the survival of patients with primary liver cancer,the COX method was used to analyze the factors affecting the prognosis of patients with primary liver cancer.Results:Compared with the control group,the relative expression level of TMED5 mRNA in the experimental group was higher(P<0.05),and the expression level of miR-652-3P was lower(P<0.05);there was a negative correlation between miR-652-3P and TMED5 in primary liver cancer tissues(P<0.05);miR-652-3P and TMED5 were related to TNM stag-ing,lymph node metastasis,lymphatic and vascular space invasion(P<0.05),but not related to the patient's age,gender,tumor size and degree of differentiation(P>0.05);Kaplan-Meier method showed that the average survival time of those with high expression of miR-652-3P was significantly longer than those with low expression of miR-652-3P,and the average survival time of those with low expression of TMED5 was significantly longer than those with high expression of TMED5
