摘要
以我国五个黄花菜主要产区山西大同、湖南祁东、甘肃庆阳、宁夏盐池、陕西大荔的黄花菜干制品为研究对象,在进行多糖提取工艺取得黄花菜精制多糖(DPH)粉末样品后,采用MTT法,测定DPH对人宫颈癌细胞HeLa、人永生化表皮细胞HaCaT、人肝癌细胞HepG2和人结肠癌细胞CaCO_(2)的生长抑制作用,MTT结果表明,五产区DPH均对试验细胞有抑制作用,并且随浓度增加而增强,其中庆阳DPH对HeLa、CaCO_(2)和HaCaT的抑制作用最佳,祁东DPH对HepG2的抑制作用最佳。采用LPS刺激小鼠单核巨噬细胞白血病细胞RAW264.7的炎症模型,通过测定NO和IL-6炎症因子浓度研究DPH的抗炎活性,结果表明与对照组相比,庆阳和盐池DPH可显著抑制IL-6产生,大同DPH可显著抑制NO产生。
The dried products of Hemerocallis citrina Baroni from Datong,Qidong,Qingyang,Yanchi,and Dali,five main Hemerocallis citrina Baroni producing areas in China were studied.After the powder samples of refined polysaccharides from Hemerocallis citrina Baroni(DPH)were obtained,the inhibitory effects of DPH on human cervical cancer cell HeLa,human immortalized epidermis cell HaCaT,human liver cancer cell HepG2 and human colon cancer cell CaCO_(2)were determined by MTT method.The MTT results showed that the inhibitory effect of DPH on the growth of human cervical cancer cell line HeLa,human immortalized epidermis cell line HaCaT,human liver cancer cell line HepG2 and human colon cancer cell line CaCO_(2).DPH in all the five producing areas had inhibitory effect on the experimental cells,and increased with the increase of concentration,among which Qingyang DPH had the best inhibitory effect on HeLa,CaCO_(2)and HaCaT,but the inhibitory effect on HaCaT was less than that of HeLa,and the inhibitory effect on HeLa was extremely significant,and Qidong DPH had the best inhibitory effect on HepG2.The inflammatory model of mouse mononuclear macrophage leukemia cell RAW264.7 stimulated by LPS was used to study the anti-inflammatory activity of DPH by measuring the concentration of NO and IL-6 inflammatory cytokines.The results showed that compared with the control group,Qingyang and Yanchi DPH could significantly inhibit the production of IL-6,and Datong DPH could significantly inhibit the production of NO.
