河北地区鸡传染性支气管炎病毒的遗传变异分析 被引量:7

摘要 为确定河北地区鸡传染性支气管炎病毒(IBV)的流行及遗传演化情况,本研究对来自河北主要养鸡地区鸡场的118份疑似IBV感染鸡的病料样品经SPF鸡胚进行病毒分离,并对传至第3代的尿囊液经PCR扩增IBV S1基因并测序后,以GenBank中的IBV疫苗株、国内外部分流行株共53株作为参考病毒,利用DNAStar 7.1软件分析分离株S1基因的同源性。采用MEGA 7.0软件中的Neighbor-Joining方法构建S1基因的遗传进化树,并分析S1基因编码的氨基酸序列。采用RDP4、SimPlot软件分析分离病毒株的重组事件。结果显示,分离到11株IBV,且其S1基因全长1614 bp~1620 bp,编码约540个氨基酸。S1基因的同源性分析结果显示,11株分离株之间的同源性为92.8%~99.9%;11株分离株与国内外53株参考株之间的同源性为73.0%~96.7%,与国内常用疫苗株M41、H120、W93、Ma5和QX的同源性为74.7%~96.1%。遗传进化分析结果显示,11株分离株与GI-19基因型(QX型)IBV属于同一分支。序列分析结果显示,11株分离株S1基因及其编码氨基酸序列与参考株相比均发生了不同程度的突变,主要突变位点在其与血清型特异性和病毒中和表位相关的高变区(aa38~aa67、aa91~aa141和aa274~aa387)内。基因重组结果显示,分离株HB.SJZ.21和HB.XT.1是两株重组病毒,HB.SJZ.21株可能是由HB.XT.9株和HB.SJZ.8株重组而成,HB.XT.1株可能是由HB.SJZ.8株和智利分离株重组而成。上述结果表明,河北地区IBV流行株为GI-19基因型,与疫苗株相比存在不同程度的变异,且分离株之间存在重组现象。本研究为河北地区鸡传染性支气管炎的流行病学调查及免疫防控提供了参考。 In order to determine the prevalence and genetic evolution of avian infectious bronchitis virus(IBV)in Hebei,the viruses were isolated from SPF chicken embryos from 118 suspected samples of chicken infected with IBV from chicken farms in Hebei province.The S1 gene of IBV was amplified from the allantoic fluid of the third generation by PCR and sequenced.Together with a total of 53 strains in GenBank including the IBV vaccine strains and the epidemic strains isolated in China and abroad as reference viruses,the homology of IBV S1 gene was analyzed by DNAStar7.1 software.The genetic evolution tree of S1 gene was constructed by Neighbor-Joining method in MEGA7.0 software.The amino acid sequence of S1gene was analyzed by MEGA7.0 software.The recombination events of isolated virus were analyzed by RDP4 and SimPlot software.The results showed that 11 samples were IBV positive.Sequence analysis of S1 gene in the positive samples showed that the total length of S1 gene was 1614bp-1620bp,encoding about 540 amino acids.The homology analysis of S1 gene showed that the homology among 11 isolates was 92.8%-99.9%.The homology between 11 isolates and 53 reference strains at home and abroad was 73.0%-96.7%,and the homology with domestic commonly used vaccine strains M41,H120,W93,Ma5and QX was 74.7%-96.1%.Genetic evolution analysis showed that 11 isolates belonged to the same branch as GI-19 genotype(QX type)IBV.The amino acid sequence analysis of S1 gene showed that the S1 gene and its amino acid sequence of 11 isolates were mutated in different degrees compared with the reference strain,and the main mutation sites were in the hypervariable regions(aa38-aa67,aa91-aa141 and aa274-aa387)related to serotype specificity and virus neutralization epitopes.The results of gene recombination showed that the isolated strains HB SJZ.21 and HB XT.1 are recombinant viruses,HB SJZ.21 strain was recombined with HB XT.9 strain and HB SJZ.8 strain.HB XT.1 strain recombined with HB SJZ.8 strain and Chilean isolate.The above results showed that th
