摘要
目的观察解郁止痛方对偏头痛抑郁共病大鼠行为学及脑组织中脑源性神经营养因子(BDNF)表达的影响,探讨解郁止痛方改善疼痛及抑郁症状的分子机制。方法将48只健康雄性SD大鼠按照随机数字表法分为空白组、模型组、尼莫地平组、解郁止痛方高剂量组、解郁止痛方中剂量组、解郁止痛方低剂量组,每组8只。除空白组外,其余组大鼠采用颈背部皮下注射硝酸甘油注射液及慢性温和不可预知刺激(实验第1,7,14,21天各干预1次)方法诱导偏头痛抑郁共病模型。实验期间,空白组和模型组大鼠每天予以纯水灌胃,尼莫地平组大鼠予以尼莫地平混悬液6 mg/kg灌胃,解郁止痛方高、中、低剂量组大鼠分别给予解郁止痛方10 g/kg、5 g/kg、2.5 g/kg(生药)灌胃,均1次/d,连续21 d。实验第1,7,14,21天造模前行机械痛阈值测定,第1,10,21天进行行为学评估;实验第21天,ELISA法检测各组大鼠血清及额叶皮质、海马、杏仁核组织中BDNF含量。结果实验第7,14,21天造模前,模型组大鼠机械痛阈值均明显低于空白组(P均<0.05),尼莫地平组及解郁止痛方各组均明显高于模型组(P均<0.05)。实验第10,21天,模型组大鼠糖水偏好百分率均明显低于空白组(P<0.05),尼莫地平组及解郁止痛方各组均明显高于模型组(P均<0.05)。实验第10、21天,模型组大鼠强迫游泳不动时间、新奇抑制摄食潜伏时间均明显长于空白组(P均<0.05);实验第10天,尼莫地平组及解郁止痛方各组大鼠强迫游泳不动时间和解郁止痛方中剂量组大鼠新奇抑制摄食潜伏时间均明显短于模型组(P均<0.05);实验第21天,尼莫地平组、解郁止痛方中剂量组大鼠强迫游泳不动时间、新奇抑制摄食潜伏时间及解郁止痛方高剂量组大鼠强迫游泳不动时间均明显短于模型组(P均<0.05)。实验第21天干预结束后,与空白组比较,模型组大鼠血清及额叶皮质、海马组织中BDNF含量均明
Objective It is to observe the effect of Jieyu Zhitong decoction on the behaviors and brain-derived neurotrophic factor(BDNF)expression in cerebral tissue of rats with migraine and depression,and to explore the molecular mechanism of this decoction in improving pain and depression symptoms.Methods According to the random number table method,48 healthy male SD rats were divided into blank group,model group,nimodipine group,high-dose Jieyu Zhitong decoction group,medium-dose Jieyu Zhitong decoction group,and low-dose Jieyu Zhitong decoction group,8 rats in each group.Except for the blank group,the rats in the other groups were treated with nitroglycerin injection by subcutaneous injection on the neck back and chronic mild and unpredictable stimulation(intervention for once on the 1st,7th,14th,and 21st days of the experiment)to induce the models of migraine-depression comorbidity.During the experiment,the rats in the blank group and the model group were gavaged with pure water every day,the rats in the nimodipine group were gavaged with 6 mg/kg nimodipine suspension,and the rats in the high-dose,medium-dose,and low-dose Jieyu Zhitong decoction groups were respectively givenJieyu Zhitong decoction 10 g/kg,5 g/kg,2.5 g/kg(crude medicine)by gavage,all once a day for 21 days.On the 1st,7th,14th,and 21st days of the experiment,the mechanical pain threshold was detected before the models were established,and the behavioral evaluation was performed on the 1st,10th,and 21st days of the experiment;on the 21st day of the experiment,the content of BDNF in serum,frontal cortex,hippocampus and amygdala of rats in each group was detected by ELISA.Results Before modeling on the 7th,14th,and 21st days of the experiment,the mechanical pain thresholds of the rats in the model group were significantly lower than those in the blank group(all P<0.05),and the thresholds in the nimodipine group and the Jieyu zhitong decoction groups were significantly higher than those in the model group(all P<0.05).On the 10th and 21st day of the experim
