摘要
目的:运用网络药理学研究方法探讨逍遥散治疗经前期综合征的作用机制。方法:在中药系统药理学数据库与分析平台(TCMSP)和中药分子作用机制的生物信息学分析工具中筛选并检索逍遥散中药物核心成分和靶点;并通过相关数据库检索经前期综合征疾病靶点;利用韦恩图得到逍遥散和疾病的共同靶点;借助Cytoscape软件描绘成分-疾病-靶点图;在STRING平台上进行蛋白质-蛋白质相互作用(PPI)分析;最后利用DAVID网站将药物及疾病之间交集的靶点进行基因本体(GO)富集分析和京都基因和基因组百科全书(KEGG)富集分析。结果:逍遥散八味中药共筛选出162种成分和2618个预测靶点,经前期综合征靶点768个;韦恩图得到共同靶点21个;得到43条生物过程,涉及信号转导、RNA聚合酶Ⅱ的转录起始、IκB激酶/核因子κB信号的负调控、凋亡过程的负调控等;同时筛选得到17条信号通路,包括HIF-1信号通路等。结论:逍遥散可能通过激素调节,调节下丘脑-垂体-肾上腺轴,抗抑郁,调节神经递质等途径治疗经前期综合征。
Objective:To explore the underlying mechanism of Xiaoyao Powder in the treatment of premenstrual syndrome based on network pharmacology.Methods:The core components and targets of Xiaoyao Powder were retrieved from the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)and Bioinformatics Analysis Tool for Molecular Mechanism of Traditional Chinese Medicine(BATMAN-TCM).The targets of the premenstrual syndrome were predicted by relevant databases.Venn diagram was used to obtain the common targets of Xiaoyao Powder and the disease.Cytoscape was used to construct the“component-target-disease”network.STRING was used for the analysis of protein-protein interaction(PPI).The common targets of Xiaoyao Powder and the disease were analyzed by Gene Ontology(GO)enrichment and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment on DAVID.Results:A total of 162 active components and 2618 predicted targets of Xiaoyao Powder and 768 targets related to premenstrual syndrome were obtained.Twenty-one common targets were obtained through Venn diagram.Forty-three biological processes were screened out,involving signal transduction,transcription initiation of RNA polymerase Ⅱ,negative regulation of I-κB kinase/NF-κB signaling,and negative regulation of apoptosis.Meanwhile,17 signaling pathways were screened out,including the HIF-1 signaling pathway.Conclusion:Xiaoyao Powder may treat premenstrual syndrome by regulation of hormones,hypothalamic-pituitary-adrenal axis,and neurotransmitters,and anti-depression.
