摘要
目的探讨木犀草素对顺铂诱导乳腺癌细胞凋亡的增敏作用。方法使用3 mg·L^(-1)顺铂处理培养乳腺癌细胞MDA-MB-231,构建细胞凋亡模型,作为模型组。在模型组的基础上分别采用低(5μg·mL^(-1))、中(10μg·mL^(-1))、高(20μg·mL^(-1))剂量的木犀草素处理细胞,作为木犀草素低、中、高剂量组;空白组的MDA-MB-231细胞以等量生理盐水干预。细胞计数试剂盒-8(CCK-8)法检测细胞增殖情况,流式细胞术检测细胞凋亡情况,Transwell检测细胞迁移情况,蛋白质印迹(Western blot)法检测细胞蛋白表达水平。结果CCK-8实验中空白组、模型组、木犀草素低、中、高剂量组的光密度(OD)值分别为1.48±0.26,1.18±0.13,0.74±0.07,0.66±0.06和0.42±0.04;5组细胞凋亡率为(5.67±0.33)%,(8.48±0.93)%,(18.85±1.22)%,(25.37±2.74)和(37.64±3.11)%;5组细胞迁移数量分别为(275.98±39.93),(200.67±28.76),(143.89±19.67),(115.67±20.68)和(83.59±7.11)个;这5组Cyclin D1蛋白的相对表达水平0.99±0.09,0.32±0.02,0.95±0.09,0.90±0.09和0.89±0.09,B细胞淋巴瘤/白血病-2(Bcl-2)蛋白的相对表达水平0.95±0.09,0.88±0.08,0.54±0.04,0.42±0.04和0.30±0.03,基质金属蛋白酶2(MMP2)蛋白的相对表达水平0.90±0.09,0.82±0.08,0.55±0.05,0.47±0.04和0.27±0.02。以上指标,模型组与低、中、高剂量组相比,差异均有统计学意义(均P<0.05)。结论木犀草素降低顺铂诱导的乳腺癌细胞增殖、迁移和侵袭,促进细胞凋亡。
Objective To investigate the sensitizing effect of luteolin on cisplatin-induced apoptosis of breast cancer cells.Methods MDA-MB-231 breast cancer cells were treated with 3 mg·L^(-1) cisplatin to construct the apoptosis model as the model group.On the basis of the model group,the cells were treated with low(5μg·mL^(-1)),medium(10μg·mL^(-1))and high(20μg·mL^(-1))doses of luteolin as low,medium and high dose groups of luteolin;mDA-MB-231 cells in the blank group were intervened with the same amount of normal saline.Cell counting kit-8(CCK-8)was used to detect cell proliferation,flow cytometry was used to detect cell apoptosis,and Transwell was used to detect cell migration.The protein expression was detected by Western blot.Results The optical density(OD)values of the blank group,the model group,and the low,medium,and high dose groups of luteolin in CCK-8 experiment were 1.48±0.26,1.18±0.1,0.74±0.07,0.66±0.06,and 0.42±0.04,respectively.The apoptosis rate was(5.67±0.33)%,(8.48±0.93)%,(18.85±1.22)%,(25.37±2.74)%and(37.64±3.11)%in 5 groups.The number of cell migration in the five groups was 275.98±39.93,200.67±28.76,143.89±19.67,115.67±20.68 and 83.59±7.11,respectively.The relative expression levels of Cyclin D1 protein in these five groups were 0.99±0.09,0.32±0.02,0.95±0.09,0.90±0.09 and 0.89±0.09,respectively.The relative expression levels of B-cell lymphoma leukemia(Bcl-2)were 0.95±0.09,0.88±0.08,0.54±0.04,0.42±0.04 and 0.30±0.03,respectively.The relative expression levels of matrix metalloproteinase-2(MMP-2)protein were 0.90±0.09,0.82±0.08,0.55±0.05,0.47±0.04 and 0.27±0.02.Compared model group with low,medium and high dose group,the difference of the above indicators was statistically significant(all P<0.05).Conclusion Luteolin reduces cisplatin-induced breast cancer cell proliferation,migration and invasion.
