基于高通量测序分析传统轮作对西洋参参田土壤细菌群落结构影响 被引量:1

Analysis of Bacterial Community Structure of Traditional Cropping Rotation on ginseng field soil Based on High-throughput Sequencing

摘要 目的研究传统轮作对西洋参参田土壤细菌群落结构的影响,为研究西洋参参田土壤改良及制定合理轮作提供依据。方法西洋参采收后大田土(A组)、轮作1年大田土(B组)、轮作2年大田土(C组)、未种植过西洋参的大田土(D组)为研究对象,采用Illumina Miseq高通量测序技术分析了土壤细菌群落结构的变化。结果12个土壤样品经16S测序共获得有效序列480148条,用于物种分类的OTU有3788个,涵盖33门,88纲,299目,390科,755属。在属水平上,各组细菌群落组成及相对丰度差别很大,其中鞘氨醇单胞菌属(Sphingomonas)和芽单胞菌属(Gemmatimonas)可能是西洋参生长的有益菌,随着传统轮作时间的增加,其丰度渐次增加。结论新茬大田和传统轮作地老参田土壤中微生物组成和结构均具有显著性差异,研究结果对于探究西洋参参田土壤改良种植及传统轮作土壤微生物变化具有理论指导作用,为建立西洋参与主要传统农作物的合理轮作制度提供实验数据。 Objective The effects of traditional cropping rotation on soil bacterial community structure of old ginseng field were studied to provide basis for soil improvement and rational cropping rotation of old ginseng field.Methods The changes of bacterial communities structure in new crop field after harvesting(group A),traditional 1-year rotation(group B),traditional 2-year rotation(group C),in uncultivated soil(group D)were analyzed by using the Illumina MiSeq high-throughput sequencing technology.Results A total of 636654 effective sequences and 8422 OTUs were obtained from nine samples based on high-throughput sequencing of the 16S gene.Bacterial species detected in these samples covered 42 phyla,106 classes,180 orders,158 families,and 246 genera.At the genera level,the composition and relative abundance of the bacterial communities in each groups made a great different.among them Sphingomonas and Gemmatimonas may be the beneficial bacterium for the growth may be the key bacteria causing root rot of Panax quinquefolium and should be focused in the further research,and their abundance increased gradually with the increase of traditional rotation time.Conclusion There are significant differences in bacterial community compositions from the soil microbial composition and structure between the new crop field and the traditional rotation field.The study provides theoretical guidance for exploring the soil microbial changes in the improved cultivation of the old ginseng field and the traditional rotation field,and provided experimental data for the establishment of a rational cropping system for western participation in the main traditional crops.
