泄泻肾阳亏虚证小鼠模型的建立与验证 被引量:2

Model Building and Validation of Diarrhea Mice with Kidney-yang Depletion Syndrome

摘要 目的探索建立泄泻肾阳亏虚证小鼠模型的方法。方法模型建立:将20只雄性昆明小鼠随机分为空白组和模型组,每组10只。模型组小鼠给予腺嘌呤混悬液50 mg/(kg·d)灌胃,每日1次,连续14日;从第8日起给予番泻叶水煎液10 g/(kg·d)灌胃,每日1次,连续7日。空白组小鼠给予0.4 ml无菌水灌胃,每日1次,连续14日。观察两组小鼠一般行为学,分别于造模第1、5、9、13日时测量小鼠体质量、肛温,计算腹泻指数,造模第14日结束时检测血清中环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)含量,计算cAMP/cGMP值。模型验证:将另外30只小鼠随机分为正常组、模型对照组和四神丸组,每组10只。模型对照组和四神丸组按照上述方法造模,造模成功后四神丸组小鼠给予四神丸水煎液5 g/(kg·d)灌胃,每日2次,连续7日;正常组与模型对照组给予0.35 ml无菌水灌胃,每日2次,连续7日。比较各组小鼠一般行为学改变,干预第1、3、7日时体质量、肛温、腹泻指数,以及干预7日后血清中cAMP、cGMP含量及cAMP/cGMP值。结果造模结果显示,空白组小鼠精神状态及自主活动情况均正常,粪便柔软成形,干湿适中,肛周洁净。模型组小鼠精神状态欠佳,体凉尾冷,爪甲颜色较浅,毛发稀疏无光泽,粪便稀软。与空白组同时间比较,模型组小鼠造模第9、13日时体质量降低,造模第5、9、13日时小鼠肛温降低(P<0.01)。模型组小鼠腹泻指数随着时间延长而逐渐升高,呈时间依赖性;模型组小鼠血清cAMP含量降低,cGMP含量升高,且cAMP/cGMP值降低(P<0.01)。模型验证结果显示,四神丸组小鼠精神状态较模型组有所恢复,活动灵活,皮毛光泽度提高,爪甲颜色恢复,粪便稀溏现象减轻。与模型对照组同时间比较,在治疗第3、7日时,四神丸组腹泻指数均显著降低(P<0.05)。给药7日后小鼠血清cAMP、cGMP水平及cAMP/cGMP值差异均无统计学意义(P>0.05)。结论腺嘌呤联合番泻叶可成 ObjectiveTo explore the modeling method of diarrhea mice with kidney-yang depletion syndrome.MethodsModel building stage:Twenty male Kunming mice were randomly divided into the blank group and modeling group,with 10 mice in each group.Mice in the modeling group were given daily gavage of 50 mg/(kg·d)adenine suspension for 14 days.From the 8th day onwards,the modeling group was gavaged with 10 g/(kg·d)of Fanxieye(Folium sennae)decoction,once a day for 7 days.Mice in the blank group were given 0.4 ml sterile water daily by gavage for 14 days.The general behavior of the mice in both groups was observed,while the body weight and rectal temperature were measured,and the diarrhea index was calculated on the 1st,5th,9th,and 13th day of modeling.At the end of the 14th day of modeling,the serum content of cyclic adenosine monophosphate(cAMP)and cyclic guanosine monophosphate(cGMP)were tested,and the cAMP/cGMP value was calculated.Model validation stage:the remaining 30 mice were randomly assigned to the normal group,model control group and Sishen Pill(四神丸)group,with 10 mice in each group.The model was built in the model control group and Sishen Pill group with the above mentioned method.After successful modeling,mice in the Sishen Pill group were gavaged with 5 g/(kg·d)of Sishen Pill decoction,twice a day for 7 days.Mice in the normal and model control groups were given 0.35 ml sterile water by gavage,twice a day for 7 days.Mice were tested for general behaviour,and body weight,rectal temperature,and diarrhea index were assessed on the 1st,3th,and 7th day.The serum content of cAMP and cGMP and the cAMP/cGMP value were assessed after 7 days.ResultsDuring the model building stage,the mental state and autonomous activity of the mice in the blank group were normal,and the perianal region was clean with soft,formed and moderate watery stool.Mice in the modeling group were in poor mental state,with cool body and cold tail,light nails,sparse and dull hair,and soft feces.Compared to those in the normal group,the body wei
