摘要
动物在生长发育不同阶段组织器官的代谢水平有所差异,线粒体DNA(mtDNA)的拷贝数在一定程度上反映了机体的生理状态和代谢强度。以64只大熊猫(Ailuropoda melanoleuca)粪便DNA为材料,采用实时荧光定量PCR方法,验证了肠道上皮组织中平均每个细胞mtDNA的拷贝数(N_(mpc))与月龄之间具有显著相关性(R^(2)=0.129,P=0.004),且呈现出随着月龄的增长而下降的趋势。幼龄个体的N_(mpc)远高于中、老龄个体,且个体差异很大。此外,性别对N_(mpc)也具有明显的影响,同一生长阶段的雄性比雌性更高。这些发现为进一步探究肠道上皮细胞mtDNA拷贝数与生长发育和衰老之间的关系,以及利用其作为指标来评估机体的生理状况提供了依据。
Metabolic rate varies with age in animal tissues and organs.The copy number of mtDNA reflects the physiological state and metabolic level of a tissue to a certain extent.Therefore,the copy number of mtDNA might associate with age.In this study,we quantified the average copy number of mtDNA per cell(N_(mpc))in fecal DNA representing shed epithelial cells of hind gut of 64 giant pandas(Ailuropoda melanoleuca)by using quantitative real-time PCR approach.The results showed a significant,but weak negative correlation between N mpc and age(R^(2)=0.129,P=0.004).N_(mpc) of young pandas varied greatly,but was significantly higher than that of middle-aged and old individuals.Meanwhile,it was found that gender had influence on copy number of mtDNA.N_(mpc) of males was greater than that of females at all age classes.These findings provided a basis for further probes into the relationship of N_(mpc) of epithelial cells and growth,development and aging of gut,and suggested new perspective for the use of feces to infer the physiological state of animals.
