摘要
目的分析壳多糖酶3样蛋白1(CHI3L1)在胰腺癌中的表达水平及其与患者预后的关系。方法将2019年6月至2020年8月西安交通大学第一附属医院收治的70例胰腺癌患者作为胰腺癌组,70例肺癌患者作为肺癌组,70例健康体检者作为健康体检组。以酶联免疫吸附法测定CHI3L1在三组血清中的表达水平,以免疫组化法测定CHI3L1在肺癌组织、胰腺癌癌旁组织、胰腺癌组织中的表达水平,以Western Blot法测定CHI3L1在肺癌细胞株A549和胰腺癌细胞株PANC-1中的表达水平。利用TCGA数据库分析胰腺癌患者CHI3L1表达与生存时间的关系。结果胰腺癌组、肺癌组、健康体检组的血清CHI3L1表达水平分别为(281.40±37.89)、(115.70±9.11)、(104.00±8.17)ng/mL;胰腺癌组的血清CHI3L1表达水平高于肺癌组、健康体检组,差异具有统计学意义(P<0.0001)。在胰腺癌癌旁组织和肺癌组织中,CHI3L1呈低表达,在胰腺癌组织中,CHI3L1的表达显著增加,且定位于细胞质。CHI3L1在胰腺癌细胞株PANC-1中的表达水平高于肺癌细胞株A549,差异具有统计学意义(P<0.01)。CHI3L1高表达组的生存时间短于CHI3L1低表达组,差异具有统计学意义(P<0.05)。结论CHI3L1在胰腺癌患者血清、组织和细胞中均呈高表达,高表达患者表现出较差的生存情况。
Objective To analyze the expression level of chitinase-3-like-protein-1(CHI3L1)in pancreatic cancer and its relationship with prognosis of patients.Methods A total of 70 patients with pancreatic cancer treated in the First Affiliated Hospital of Xi'an Jiaotong University from June 2019 to August 2020 were selected as pancreatic cancer group,70 patients with lung cancer as lung cancer group,and 70 healthy people as health examination group.The expression level of serum CHI3L1 in the three groups was measured by enzyme-linked immunosorbent assay,the expression level of CHI3L1 in lung cancer tissue,adjacent tissue of pancreatic cancer and pancreatic cancer tissue was measured by immunohistochemistry,and the expression level of CHI3L1 in lung cancer cell line A549 and pancreatic cancer cell line PANC-1 was measured by Western Blot.The relationship between CHI3L1 expression and survival time of pancreatic cancer patients was analyzed by TCGA database.Results The expression level of serum CHI3L1 in the pancreatic cancer group,lung cancer group and health examination group were(281.40±37.89),(115.70±9.11)and(104.00±8.17)ng/mL respectively;the expression level of serum CHI3L1 in the pancreatic cancer group was higher than that in the lung cancer group and the health examination group,and the difference was statistically significant(P<0.0001).The expression of CHI3L1 was low in lung cancer tissue and adjacent tissue of pancreatic cancer,in pancreatic cancer tissue,the expression of CHI3L1 was significantly increased and localized in the cytoplasm.The expression level of CHI3L1 in pancreatic cancer cell line PANC-1 was higher than that in lung cancer cell line A549,and the difference was statistically significant(P<0.01).The survival time of CHI3L1 high expression group was shorter than that of CHI3L1 low expression group,and the difference was statistically significant(P<0.05).Conclusion CHI3L1 is highly expressed in serum,tissue and cell of patients with pancreatic cancer,and patients with high expression show poor su
