MicroRNA-212通过调控TGF-β1/Smads信号通路对积水肾纤维化进展的影响 被引量:1

Effect of microRNA-212 regulation on the development of hydronephrosis fibrosis through TGF-beta1/Smads signal way

摘要 目的:探讨microRNA-212通过调控TGF-β1/Smads信号通路对积水肾纤维化进展的促进作用。方法:24只8周龄雄性C57BL/6J小鼠随机分为Sham组、a组、b组和c组四组,每组6只。Sham组行假手术,a组行单侧输尿管结扎阻断,b组和c组假手术后即开始分别每天静脉注射从a组小鼠提取的Exo/miroRNA-212血清和生理盐水。10 d后分别提取肾脏组织行Masson染色评估肾纤维化比例(FR),免疫组化检测α平滑肌肌动蛋白(α-SMA)的表达,Western Blot测TGF-β1、p-Smad3蛋白表达情况。结果:a组和b组FR、α-SMA的表达百分比、TGF-β1和Smad3蛋白表达量显著高于Sham组和c组(P<0.05)。结论:输尿管梗阻肾脏通过外泌体转运的microRNA-212上调TGF-β1/Smad3表达,从而促进肾纤维化的发生发展。 Objective:To study the facilitation of microRNA-212 on the process of hydronephrosis fibrosis by targeting TGF-β1/Smad.Methods:A total 24 eight-week-old male mice were randomly allocated into 4 groups(group Sham,group a,group b and group c).The mice in group a were underwent unilateral ureter obstruction.Group b and group c were injected Exo/microRNA-212 serum which extracted from mice in group a.10 days later renal tissues were harvested.Renal fibrosis assessed by Masson stain andα-SMA expression.The fibrosis related protein TGF-β1 and p-Smad3 were also measured by Western Blot.Results:Compared with those in group Sham and group c,fibrosis rate(FR)and expression ofα-SMA,TGF-β1,and Smad3 were significantly higher in group a and group b(P<0.05).Conclusion:The development of renal fibrosis was activated by exosome-delivered microRNA-212 upregulated the expression of TGF-β1/Smad3 in ureteral obstruction.
