摘要
目的探讨云南地区傣族、汉族人群糖基化终产物受体(RAGE)基因多态性与糖尿病视网膜病变(DR)的相关性。方法本研究选取昆明地区汉族人群、西双版纳地区汉族人群、西双版纳地区傣族人群各150例,每个地区民族人群包括正常对照50例,糖尿病未合并DR者50例,糖尿病合并DR者50例,分别纳入对照组、NDR组和DR组,检测RAGE基因-374T/A、2184A/G、2245G/A基因位点基因分型,比较不同地区或民族各组基因型和基因频率分布差异,分析RAGE基因多态性与糖尿病患者DR发生的关系。结果西双版纳地区傣族人群中DR组RAGE-374T/A位点A等位基因分布频率高于NDR组和对照组,T等位基因分布频率低于NDR组和对照组(P<0.05);RAGE 2184A/G位点G等位基因分布频率高于NDR组和对照组,A等位基因分布频率低于NDR组和对照组(P<0.05)。西双版纳地区傣族人群中DR组与NDR组RAGE 2245G/A位点基因型和基因频率分布比较差异无统计学意义(P>0.05),而NDR组和DR组RAGE 2245G/A位点G等位基因分布频率均低于对照组,A等位基因分布频率高于对照组(P<0.05)。昆明地区汉族人群、西双版纳地区汉族人群中三组间RAGE基因-374T/A、2184A/G、2245G/A位点基因型和基因频率分布差异无统计学意义(P>0.05)。非条件Logistic回归模型结果显示RAGE-374T/A位点TA+AA基因型、RAGE 2245G/A位点AG+GG基因型是西双版纳地区傣族糖尿病患者DR发生的危险基因型。结论云南西双版纳地区傣族人群RAGE基因-374T/A位点TA+AA基因型、2184A/G位点AG+GG基因型增加了糖尿病患者视网膜病变发生的风险。
Objective To explore the correlation between receptor for advanced glycation end product(RAGE)gene polymorphisms and diabetic retinopathy(DR)in Dai and Han populations in Yunnan.Methods A total of 150 Han individuals from Kunming,150 Han individuals from Xishuangbanna,and 150 Dai individuals from Xishuangbanna were enrolled in this study.In each region,there were 50 normal individuals as control group,50 individuals with diabetes mellitus but without DR as NDR group,and 50 individuals with diabetes mellitus and DR as DR group.The genotypes at RAGE-374T/A,2184A/G and 2245G/A were detected.The genotypes and gene frequencies in different regions or nationalities were compared.The relationship between RAGE gene polymorphisms and DR was analyzed.Results For Dai individuals from Xishuangbanna,A allele frequency at-374T/A and G allele frequency at RAGE 2184A/G were higher in DR group than in NDR group and control group,while T allele frequency at-374T/A and A allele frequency at RAGE 2184A/G were lower in DR group than in NDR group and control group(P<0.05).For Dai individuals from Xishuangbanna,genotypes and gene frequency at RAGE 2245G/A showed no statistically significant difference between DR group and NDR group(P>0.05),and G allele frequency at RAGE 2245G/A was lower in NDR group and DR group than in control group,while A allele frequency was higher in NDR group and DR group than in control group(P<0.05).For Han individuals from Kunming and Xishuangbanna,genotypes and gene frequencies at RAGE-374T/A,2184A/G and 2245G/A were similar between the three groups(P>0.05).Unconditional Logistic regression analysis showed that TA and AA genotype at RAGE-374T/A,AG and GG genotype at RAGE 2245G/A were risk genotypes for DR in Dai individuals from Xishuangbanna.Conclusion TA and AA genotype at RAGE-374T/A,and AG and GG genotype at 2184A/G can increase the risk of DR in Dai individuals with diabetes mellitus from Xishuangbanna.
