RAB1A在垂体腺瘤组织中的表达及临床意义

摘要 目的探讨RAB1A在垂体腺瘤(PA)组织中的表达及临床意义。方法收集2018年1月至2021年8月手术切除并经术后病理确诊的104例PA,根据Knosp分类法判断肿瘤侵袭性。用免疫组织化学染色法和免疫印迹法检测PA组织RAB1A的表达水平。结果104例PA中,侵袭性41例,非侵袭性63例。侵袭性PA组织RAB1A高表达率为[65.85%(27/41)]明显高于非侵袭性PA组织[36.51%(23/63);P<0.05]。侵袭性PA组织RAB1A蛋白相对表达量(2.87±0.54)明显高于非侵袭性PA组织([1.67±0.48);P<0.05]。多因素logistic回归分析显示RAB1A高表达(OR=1.908;95%CI 1.004~3.786;P=0.003)是PA侵袭性生长的独立影响因素。ROC曲线分析显示RAB1A鉴别PA侵袭性生长的曲线下面积为0.851,灵敏度、特异度分别为85.13%和79.12%。结论RAB1A的表达水平与PA的侵袭性有关,检测RAB1A可辅助评估PA的侵袭性。 Objective To investigate the expression of Ras-related protein Rab-1A(RAB1A)in pituitary adenoma(PA)tissues and its clinical significance.Methods The expression of RAB1A was detected in PA tissues obtained form 104 PA patients who underwent surgery from January 2018 to August 2021 using immunohistochemical staining and Western-bloting,respectively.The invasiveness of PA was assessed by Knosp grade.Results Of 104 PA patients,41 patients were classified as invasive PA and 63 as non invasive.The high expression rate of RAB1A in invasive PA tissues[65.85%(27/41)]was significantly higher than that in non-invasive PA tissues[36.51%(23/63);P<0.05].The relative expression of RAB1A protein in invasive PA tissues(2.87±0.54)was significantly higher than that in non-invasive PA tissues[(1.67±0.48);P<0.05].multivariate logistic regression analysis showed that high expression of RAB1A was an independent risk factor for PA invasion(OR=1.908;95%CI 1.004~3.786;P=0.003).ROC curve analysis showed a area under curve of 0.851,a sensitivity of 85.13%and a specificity of 79.12%for RAB1A expression to differentiate the invasiveness of PA.Conclusions The expression level of RAB1A is related to the invasiveness of PA,and detection of RAB1A can help evaluating the invasiveness of PA.
