推拿对坐骨神经慢性压迫损伤模型大鼠脊髓背角星形胶质细胞的影响 被引量:1

Effects of Tuina therapy on astrocytes at spinal dorsal horn in rats with chronic constriction injury

摘要 目的:观察推拿对坐骨神经慢性压迫损伤模型(CCI)大鼠脊髓背角星形胶质细胞表达的影响,探讨推拿对神经病理性疼痛(NPP)的镇痛效应及脊髓中枢机制。方法:64只SD大鼠随机分为空白组、假手术组、模型组、模型+推拿组,每组16只。采用羊肠线结扎的方法建立CCI模型;模型+推拿组造模后第4—13天,每日推拿1次,其余组均不做推拿。观察各组大鼠行为学变化及脊髓星形胶质细胞表达情况。结果:模型组大鼠造模后PWT、PWL均显著低于空白组及假手术组(P<0.01);造模后第10、14天,模型+推拿组与模型组比较,PWT、PWL显著升高(P<0.05,P<0.01);与空白组和假手术组比较,模型组大鼠造模后脊髓背角星形胶质细胞明显激活,GFAP蛋白表达量显著升高(P<0.01);模型+推拿组脊髓背角星形胶质细胞较模型组激活减少,GFAP蛋白表达量显著降低(P<0.01)。结论:推拿能够通过抑制脊髓背角星形胶质细胞激活,降低CCI造模导致的痛觉过敏和触诱发痛。 Objective:To observe the effects of Tuina on the expression of astrocytes in the spinal dorsal horn of rat model of chronic constriction injury(CCI),and to explore the Tuina spinal cord mechanism of neuropathic pain(NPP)analgesic effects.Methods:Sixty-four SD rats were randomly divided into naive group,sham group,model group and CCI+Tuina group,16 rats in each group.The model group and the Tuina group used the sheep gut ligate the sciatic nerve to establish the CCI model;the CCI+Tuina group received Tuina once a day from day 4 to day13,while the rest group didn’t have Tuina operation.The behavioral changes and the expressions of astrocytes in the spinal cord of the four groups were observed.Results:The PWT and PWL of the CCI group were significantly lower than those of the naive group and the sham group(P<0.01).On the 10th and 14th days after modeling,the PWT and PWL of the CCI+Tuina group were significantly lower than those in the CCI group(P<0.05,P<0.01).After modeling,activation of astrocytes and the expression of GFAP protein in the spinal dorsal horn were significantly increased,which was significantly different from those of the naive group and sham group(P<0.01).Compared with the CCI group,astrocytes in the spinal dorsal horn of the CCI+Tuina group had less activation,and the expression of GFAP protein was lower,which were statistically different from the model group(P<0.01).Conclusion:Tuina can inhibit hyperalgesia and tactile pain caused by CCI modeling by inhibiting activation of astrocytes in spinal dorsal horn.
