摘要
目的在细胞水平上对增液汤治疗2型糖尿病的药效作用进行拆方研究,分析其组方原理。方法采用棕榈酸诱导建立HepG2肝癌细胞、C2C12骨骼肌细胞、3T3-L1脂肪细胞的胰岛素抵抗模型,给予增液汤及其拆方组处理后,通过测定2-NBDG的荧光强度来评价各组对细胞糖摄取的影响。借助胰岛素试剂盒来研究增液汤及其拆方组对葡萄糖刺激的MIN6胰岛细胞胰岛素分泌的影响。结果增液汤中三味中药分别在不同的细胞模型上发挥促进糖摄取、胰岛素分泌的作用。在HepG2细胞上,三味药促进糖摄取的效果相差不大;在C2C12细胞上,君药玄参发挥显著促进糖摄取的作用;在3T3-L1细胞上,麦冬有显著的作用。在MIN6细胞上,君药玄参具有很好的促进胰岛素分泌的作用。结论增液汤的整体药效通过方中三味药共同发挥,君药玄参相较于其他两味药做出的贡献更大,但臣药生地、麦冬的作用也不可或缺,说明增液汤的组方依据是合理的。
OBJECTIVE To investigate the pharmacodynamic effect of ZengYe Decoction in the treatment of type 2 diabetes in vitro,and analyze the principle of prescription.METHODS The insulin resistance models of HepG2 hepatoma cells,C2C12 skelet al muscle cells and 3T3-L1 adipocytes were induced by palmitic acid.After treatment with ZengYe Decoction and its disassembled group,the effect of each group on cell glucose uptake was evaluated by measuring the fluorescence intensity of 2-NBDG.The effect of ZengYe Decoction and its disassembled group on insulin secretion of MIN6 islet cells stimulated by glucose was studied with the help of insulin kit.RESULTS The three traditional Chinese medicines in ZengYe Decoction play the role of promoting glucose uptake and insulin secretion in different cell models.On HepG2 cells,the effects of the three drugs on promoting sugar uptake were similar;On C2C12 cells,Radix Scrophulariae played a significant role in promoting sugar uptake;Ophiopogon japonicus had a significant effect on 3T3-L1 cells.On MIN6 cells,Radix Scrophulariae had a good effect on promoting insulin secretion.CONCLUSION The overall efficacy of ZengYe decoction is brought into play through the three drugs in the prescription.The monarch drug Radix Scrophulariae makes a greatercontribution than the other two drugs,but the role of the minister drug Rehmannia glutinosa and Ophiopogon japonicus is also indispensable,indicating the prescription basis of ZengYe Decoction is reasonable.
