摘要
目的探讨丙泊酚对过氧化氢诱导的大鼠肝Kupffer细胞氧化应激损伤及磷脂酰肌醇-3激酶(PI3K)/蛋白激酶B(Akt)信号通路的影响。方法从SD大鼠肝组织中分离出Kupffer细胞。用100μmol/L过氧化氢诱导Kupffer细胞建立氧化应激损伤细胞模型,并随机分为模型组、丙泊酚低浓度组、丙泊酚中浓度组、丙泊酚高浓度组和阳性对照组,另取正常Kupffer细胞作为对照组。对照组和模型组均以不含丙泊酚的细胞培养液培养,丙泊酚低、中、高浓度组分别以含5μmol/L、25μmol/L、50μmol/L丙泊酚的细胞培养液培养,阳性对照组以含0.1μmol/L盐酸右美托咪定的细胞培养液培养。培养12 h后,检测细胞活性、细胞凋亡率,细胞中乳酸脱氢酶(LDH)释放量、活性氧簇含量、过氧化氢酶(CAT)活性、PI3K和Akt磷酸化水平。结果与对照组相比,模型组Kupffer细胞活性、CAT活性、PI3K和Akt的磷酸化水平均降低,凋亡率、LDH释放量、活性氧簇含量均升高(均P<0.05);与模型组相比,阳性对照组、丙泊酚低、中浓度组Kupffer细胞活性、CAT活性、PI3K和Akt的磷酸化水平均升高,凋亡率、LDH释放量、活性氧簇含量均降低(均P<0.05),而丙泊酚高浓度组Kupffer细胞CAT活性、PI3K和Akt的磷酸化水平均降低,凋亡率、LDH释放量、活性氧簇含量均升高(均P<0.05)。结论低、中浓度(5μmol/L、25μmol/L)的丙泊酚能够减轻过氧化氢诱导的Kupffer细胞氧化应激损伤,提高细胞活性,抑制细胞凋亡,其机制可能与激活PI3K/Akt通路有关。
Objective To explore the effects of propofol on hydrogen peroxide-induced oxidative stress damage and phosphatidylinositol-3 kinase(PI3K)/protein kinase B(Akt)signaling pathway in rat liver Kupffer cells.Methods Kupffer cells were isolated from liver tissues of SD rats.The cell models of oxidative stress damage were established using Kuppfer cells induced by hydrogen peroxide of 100μmol/L,and those cells were randomly assigned to model group,low-concentration propofol group,moderate-concentration propofol group,high-concentration propofol group,or positive control group;in addition,normal Kupffer cells acted as control group.The control and model groups were cultured using cell culture solution without propofol,whereas the low-,moderate-,and high-concentration propofol groups were cultured using cell culture solution containing propofol of 5μmol/L,25μmol/L,and 50μmol/L,respectively;moreover,the positive control group was cultured with cell culture solution containing dexmedetomidine hydrochloride of 0.1μmol/L.Cytoactivity and apoptosis rate,as well as releasing amount of lactate dehydrogenase(LDH),reactive oxygen species content,catalase(CAT)activity,and phosphorylation levels of PI3K and Akt in the cells were detected 12 hours after culture.Results Compared with the control group,the model group exhibited declines in Kupffer cell activity,CAT activity,and phosphorylation levels of PI3K and Akt,whereas increases in apoptosis rate,LDH releasing amount,reactive oxygen species content(all P<0.05);compared with the model group,the positive control group,and the low-and moderate-concentration propofol groups showed increases in Kupffer cell activity,CAT activity,and phosphorylation levels of PI3K and Akt,whereas declines in apoptosis rate,LDH releasing amount,and reactive oxygen species content(all P<0.05);however,the high-concentration propofol group yielded declines in CAT activity of Kupffer cells and phosphorylation levels of PI3K and Akt,whereas increases in apoptosis rate,LDH releasing amount,and reactive o
