摘要
目的:研究红参皂苷Rh2抑制人涎腺腺样囊性癌ACC-M细胞体外嗜神经增殖的影响。方法:将人涎腺腺样囊性癌ACC-M细胞分别与浓度为40μg/ml、80μg/ml、160μg/ml、240μg/ml红参皂苷Rh2和SD大鼠面神经培养液共培养,培养24h、48h、72h。应用MTT比色法检测红参皂苷Rh2抑制人涎腺腺样囊性癌ACC-M细胞体外嗜神经增殖的抑制生长率,并用ELISA实验定性定量检测红参皂苷Rh2抑制人涎腺腺样囊性癌ACC-M细胞体外嗜神经增殖的神经生长因子(NGFs)的表达。结果:40~240μg/ml的红参皂苷Rh2均可显著抑制人涎腺腺样囊性癌ACC-M细胞体外嗜神经的增殖。ELISA实验结果表明红参皂苷Rh2可以使SD大鼠面神经培养液当中的NGFs的表达水平显著下降(P<0.05)。结论:红参皂苷Rh2通过抑制SD大鼠面神经培养液当中的神经生长因子的表达而具有抑制人涎腺腺样囊性癌ACC-M细胞体外嗜神经增殖的能力。
Objective:To study the effect of red ginseng saponin Rh2 on the proliferation of ACC-gland adenoid cystic carcinoma in ACC-M cells.Methods:ACC-M cells of human adenoid cystic carcinoma were cocultured with concentrations of 40μg/ml,80μg/ml,160μg/ml,240μg/ml red ginseng saponin Rh2 and SD rats for 24,48 and 72 hours.MTT colorimetric method was used to measure the inhibitory growth rate of red ginseng saponin Rh2 in ACC-M cells,and the expression of nerve growth factors(NGFs)inhibited by red ginseng saponin Rh2 in ACC-M cells by ELISA.Results:40~240μg/ml red ginseng saponin Rh2 inhibited the proliferation of ACC adenoid ACC-M cells in vitro.ELISA results showed that red ginseng saponin Rh2 can significantly reduce the expression level of nerve growth factors in the facial nerve culture medium of SD rats(P<0.05).Conclusion:Red ginseng saponin Rh2 inhibited the nerve addiction proliferation ability of human salivary adenoid cystic carcinoma ACC-M cells by inhibiting the expression of nerve growth factor in SD rat facial nerve culture medium.
