辛伐他汀对小鼠胆囊结石形成及瘦素的影响

摘要 目的分析辛伐他汀对胆囊结石形成的预防效果及对瘦素(Leptin)的影响。方法饲养50只雌性C57BL/6J小鼠进行前瞻性研究,8周龄,SPF级,给予适应性喂养2周后,采用随机数字表法分为将其分为4组:基础饲料对照组(G-Control组,n=10)、基础饲料他汀药物干预组(G-Satins组,n=10),高脂饲料对照组(L-Control组,n=15),高脂饲料他汀药物干预组(L-Satins组,n=15)。G-Control组、G-Satins组饲喂基础饲料,L-Control组、L-Satins组饲喂高脂饲料。于饲养20 d后,于内眦静脉取血,检测L-Control组、L-Satins组小鼠血清总胆固醇水平,如已造模成功为高血脂小鼠,处死L-Control组、L-Satins组随机各5只小鼠,肉眼及偏光显微镜下观察小鼠的胆囊结石形成情况。于构建高血脂小鼠但尚未形成胆囊结石时,G-Satins组、L-Satins组小鼠给予辛伐他汀(20 mg/kg)羧甲基纤维素钠混悬液灌胃,共4周,每日灌胃1次,对照组给予等体积的空白羧甲基纤维素钠溶液灌胃;上述两组小鼠造模成功直至给药结束期间,持续给予高脂饲料。再饲养4周后,检测血清Leptin、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)含量。结果成石率:G-Control组、G-Satins组小鼠未见胆囊结石,L-Control、L-Satins组出现结石,L-Control组成石率较L-Satins组高,差异有统计学意义(P<0.05)。血清TC、LDL-C、Leptin含量从低到高排列为G-Satins组、G-Control组、L-Satins组、L-Control组,差异均有统计学意义(P<0.05);血清HDL-C含量的结果从高到低排列为L-Satins组、G-Control组、G-Satins组、L-Control组,差异均有统计学意义(P<0.05)。结论辛伐他汀可能通过抑制Leptin的释放,升高血清HDL-C含量及减低血清LDL-C含量,导致胆汁胆固醇沉积减少,使胆汁中胆固醇含量减少,从而降低胆囊胆固醇结石成石率。 Objective To analyze the preventive effect of simvastatin on gallstone formation and Leptin.Methods Fifty female C57BL/6J mice,aged 8 weeks,SPF grade,were fed adaptive-feeding for 2 weeks,and divided into four groups by random number table method:basal diet control group(G-Control group)(n=10),basal diet statin intervention group(G-Satins group)(n=10),high fat diet control group(L-Control group)(n=15),high fat diet statin intervention group(L-Satins group)(n=15).The pigs in G-Control group and G-Satins group were fed basal diet,while the pigs in L-Control group and L-Satins group were fed high-fat diet.After 20 days of feeding,blood was collected from inner canthal vein to detect the serum total cholesterol level of mice in L-Control group and L-Satins group.If hyperlipidemia mice were successfully modeled,5 mice in L-Control group and 5 mice in L-Satins group were killed randomly,and the gallstone formation of mice was observed under naked eyes and polarized light microscope.When hyperlipidemia mice were constructed but gallstones were not formed,mice in G-Satins group and L-Satins group were given simvastatin(20 mg/kg)carboxymethyl cellulose sodium suspension by intragastric administration,once a day for 4 weeks,while mice in control group were given blank carboxymethyl cellulose sodium solution by intragastric administration.The mice in the above two groups were given high-fat diet until the end of administration.After another 4 weeks of feeding,serum Leptin,total cholesterol(TC),high-density lipoprotein cholesterol(HDL-C)and low-density lipoprotein cholesterol(LDL-C)levels were detected.Results Stone formation rate:no gallstones were found in the G-Control group and G-Satins group,but stones were found in the L-Control and L-Satins groups,and stone formation rate of L-Control was higher than that of L-Satins group,and the difference was statistically significant(P<0.05).The levels of serum TC,LDL-C,and Leptin were ranked from low to high as G-Satins group,G-Control group,L-Satins group,and L-Control group,and
