表达传染性喉气管炎病毒gB基因的重组鸡痘病毒的免疫效力评价 被引量:1

Immune efficacy of recombinant fowlpox virus expressing gB gene of infectious laryngotracheitis virus

摘要 为了评价表达传染性喉气管炎病毒(ILTV)g B基因的重组鸡痘病毒疫苗(rFPV-ILTV g B)的免疫效力,首先将49日龄SPF鸡随机分成5组,以羽毛囊涂抹法攻毒1×10^(2)、1×10^(3)、1×10^(4)EID_(50)鸡痘病毒(FPV)10^(2)株,并设立1组1×10^(3)EID_(50)刺种攻毒对照组和1组PBS对照组,建立鸡痘攻毒模型。接着,将21日龄SPF鸡随机分组,分别以翅部刺种和颈部皮下注射方式免疫2×10^(4)PFU的rFPV-ILTV gB,免疫后第28天,分别使用FPV10^(2)株和ILTV I19株攻毒,统计发病死亡情况。结果表明,FPV10^(2)株的最小感染剂量为1×10^(3)EID_(50),最佳攻毒方式为羽毛囊涂抹;rFPV-ILTV g B经翅部刺种免疫后对ILTV I19和FPV 10^(2)株的攻毒保护率分别为100%和90%,经颈部皮下注射方式免疫后对ILTV I19和FPV 10^(2)株的攻毒保护率分别为90%和40%。本研究建立了可靠的FPV攻毒模型,并证实rFPV-ILTV g B重组活疫苗的翅部刺种免疫对ILTV和FPV攻毒均能提供很好的免疫保护作用。 The purpose of this study was to evaluate the immune efficacy of the recombinant fowlpox virus(FPV)expressing g B gene of infectious laryngotracheitis virus(r FPV-ILTV g B).Firstly,49-day-old SPF chickens were randomly divided into 5 groups in which 3 groups were challenged by FPV 102 strain at the doses of 1×10^(2),1×10^(3)and 1×10^(4)EID_(50) via smearing feather bursa on the back,1 group was challenged by FPV 102 strain at the dose of 1×10^(3)EID_(50) via stab-inoculation on the wing,and 1 group was set up as PBS control for establishing FPV pathological model.Then 21-day-old SPF chickens were immunized with2×10^(4)PFU of r FPV-ILTV g B by subcutaneous injection of neck or stab-inoculation on the wing,respectively.The immunized chickens were challenged by FPV 102 strain or ILTV I19 strain respectively 28 days after immunization,and the morbidity and mortality were counted.The result showed that the minimum infection dose of FPV 102 strain was 1×10^(3)EID_(50),and the best route to challenge the virus was smearing feather bursa on the back.The protective rates of r FPV-ILTV g B were 100%against ILTV I19 strain and 90%against FPV102 strain by stab inoculation on the wing,the protective rates were 90%and 40%by subcutaneous injection of neck,respectively.In a word,a reliable FPV pathological model had been established,and the combination live vaccine of r FPV-ILTV g B could provide good immune protection against ILTV and FPV challenge in immunized SPF chickens via stab-inoculation on the wing.
