建兰种子无菌萌发技术研究 被引量:1

Research on Aseptic Germination Technology of Cymbidium ensifolium Seeds

摘要 以赣南地区野生建兰蒴果为材料,研究了不同培养基对种子无菌萌发的影响以及NAA浓度对无菌芽生根的影响,并对建兰种子无菌萌发过程进行了观察。结果表明:添加外源植物生长调节剂6-BA、NAA以及适宜浓度的蔗糖能显著提高建兰种子的萌发率,建兰种子无菌萌发最适培养基为1/2 MS+8 g/L琼脂粉+60 g/L蔗糖+2.0 mg/L NAA+0.4 mg/L 6-BA+1 g/L活性炭;当0.8 mg/L NAA与0.4 mg/L 6-BA组合时,无菌芽生根率最高,在90 d时最高可达80.50%;通过观察建兰种子无菌萌发过程发现,建兰种子在播种后到形成根状茎分枝大约要经历250 d。 The fruits of wild Cymbidium ensifolium in southern Jiangxi were used as materials,the effects of different media on the aseptic germination of seeds and the NAA concentration on the rooting of aseptic buds were studied,and the aseptic germination process of Cymbidium ensifolium seeds was observed.The results showed that the addition of exogenous plant growth regulators(6-BA,NAA)and sucrose with appropriate concentration could significantly improve the germination rate of Cymbidium ensifolium seeds,the optimum medium for aseptic germination of Cymbidium ensifolium seeds was1/2 MS+8 g/L agar powder+60 g/L sucrose+2.0 mg/L NAA+0.4 mg/L 6-BA+1 g/L activated carbon.When 0.8 mg/L NAA and 0.4 mg/L 6-BA were combined,the rooting rate was the highest,up to 80.50%at 90 days.By observing the process of aseptic germination of Cymbidium ensifolium seeds,it was found that it took about 250 days for Cymbidium ensifolium seeds to form rhizome branches after sowing.
