天津野生山樱花组织培养与再生体系的建立 被引量:1

摘要 本试验以天津发现的野生山樱花(Cerasus serrulate L.)幼嫩茎段为试材,设置不同的激素配比、培养基支撑物和移栽基质,筛选适宜不定芽分化增殖、诱导生根和移栽炼苗的最优条件,建立野生山樱花高效组培再生体系。结果表明:丛生芽增殖的最佳培养基为MS+0.50 mg·L^(-1)TDZ+1.00 mg·L^(-1)GA;+0.1 mg·L^(-1)NAA,增殖系数可达3.96,玻璃化率最低(6%)且玻璃化程度轻,叶片嫩绿明显增大,芽苗生长健壮。最佳生根诱导培养基为以水苔为支撑物的1/2MS+0.10 mg·L^(-1)IBA+0.10 mg·L^(-1)NAA,生根率最高,可达94.59%,单株不定根数为3条,单株侧根数高达10条。最佳移栽基质为蛭石+珍珠岩+草炭(体积比2∶1∶1),移栽成活率最高为98.21%。最适宜的炼苗时间为水苔支撑生根闭瓶炼苗5 d。本研究建立了天津野生山樱花组织培养与植株再生体系,为北方山樱花野生资源在妥善保护的基础上开发应用提供了依据。 The young stem segments of wild Cerasus serrulate L. founded in Tianjin were used as materials to study the optimal conditions for adventitious bud differentiation and proliferation, root induction and seedling hardening by setting different hormone ratios, medium supports and transplanting substrates, so as to establish an efficient tissue culture and regeneration system of C. serrulate L. The results showed that the optimal medium for proliferation of clustered buds was MS+0.50 mg·L^(-1)TDZ+1.00 mg·L^(-1)GA;+0.1 mg·L^(-1)NAA;in these conditions, the proliferation coefficient was 3.96, the vitrification rate was lower as 6% with light vitrification degree, the leaves were green and enlarged obviously, and the seedlings grew well. The optimal medium for root induction was 1/2 MS+0.10 mg·L^(-1)IBA+0.10 mg·L^(-1)NAA with sphagna as support, under these conditions, the highest rooting rate could be obtained as 94.59%, and the number of lateral roots per plant was as high as 10. The optimal transplanting substrate was the mixture of vermiculite, perlite and peat at the ratio of 2∶1∶1, and the highest survival rate was 98.21%. The suitable time for seedling hardening was 5 days in closed bottle with sphagna as rooting support. The establishment of tissue culture and regeneration system could provide references for C. serrulate wild resources protect, development and application.
