摘要
目的 观察方格星虫三肽对人黑色素瘤细胞A375细胞增殖的影响。方法 CCK-8法观察方格星虫三肽对A375细胞生长的影响;平板克隆实验观察其对A375细胞克隆形成能力的影响;transwell细胞迁移实验检验其对A375细胞迁移的影响;活性氧(ROS)、乳酸脱氢酶(LDH)实验检测其对A375细胞ROS和LDH含量的影响。结果 方格星虫三肽可抑制A375细胞增殖、克隆形成、细胞迁移、LDH释放,增加ROS含量(P<0.05或0.01)。结论 方格星虫三肽对A375细胞增殖有明显抑制作用。
Objective To investigate the effect of Sipunculus nudus Linnaeus-derived tripeptide on proliferation of human melanoma A375 cells. Methods The proliferation, clonality, migration, and reactive oxygen species(ROS) and lactate dehydrogenase(LDH) levels in A375 cells were respectively detected by CCK-8, plate clone formation, transwell, DCFH-DA,and ELISA after pretreatment with the tripeptide. Results The tripeptide decreased proliferation, clone formation, migration,and LDH level in A375 cells, and increased ROS content(P<0.05 or 0.01). Conclusion The tripeptide derived from Sipunculus nudus Linnaeus can significantly inhibit the proliferation of A375 cells.
