摘要
【目的】氮磷钾是植物生长过程中不可或缺的元素,从菜园土壤中分离解磷菌、解钾菌、固氮菌,为制备微生物肥料提高农作物生长奠定基础。【方法】采用梯度稀释涂布法进行初筛,摇瓶培养复筛,分别测定发酵液中可溶性磷、有效态钾和总氮含量,拟筛选出高效解磷菌、解钾菌、固氮菌。【结果】通过试验筛选出高效解磷、解钾、固氮菌各1株,通过生理生化特征和16S rDNA序列分析进行分类鉴定出WP3-11为阿氏芽孢杆菌(Priestia aryabhattai),JK13为玉米微杆菌(Microbacterium zeae),GN7为辣椒芽孢杆菌(Bacillus zanthoxyli)。【结论】试验结果为高效生产氮磷钾菌肥,促进植物生长提供了参考。
[Objective]Nitrogen,phosphorus and potassium are indispensable elements in the process of plant growth.Phosphorus bacteria,potassium-dissolving bacteria and nitrogen-fixing bacteria were isolated from vegetable garden soil,which laid the foundation for preparing microbial fertilizer to improve crop growth.[Method]The gradient dilution coating method was used for primary screening,shake flask culture and secondary screening;the contents of soluble phosphorus,available potassium and total nitrogen in fermentation broth were determined to high phosphorus-solubilizing,potassium-solubilizing and nitrogen-fixing.[Results]The results showed that one strain with high phosphorus-solubilizing,potassium-solubilizing and nitrogen-fixing efficiency was screened out,and identified by physiological and biochemical characteristics and 16S rDNA sequence analysis:WP3-11 was Priestia aryabhattai,JK13 Microbacterium zeae,and GN7 Bacillus zanthoxyli.[Conclusion]The experimental results provided a reference for efficient production of nitrogen,phosphorus and potassium bacterial fertilizer in promoting plant growth.
