摘要
目的比较不同品牌核酸提取试剂盒在自建新型冠状病毒核酸检测中的性能差异,为临床实验室自建新型冠状病毒核酸检测系统提供参考依据。方法将第3方新型冠状病毒核酸检测质控品稀释成4种浓度,采用上海之江生物科技股份有限公司(A)、重庆中元生物技术有限公司(B)、广州和信生物科技有限公司(C)及杭州博日科技有限公司(D)4个不同厂家核酸提取试剂组成的4种自建检测系统进行检测,比较其阳性检出率及批内重复性。结果在不同浓度样本中,D品牌提取试剂盒提取的样本结果阳性率最高,靶基因检出率最高;B品牌扩增结果的Ct值最小,提取产物浓度最高。结论不同品牌核酸提取试剂的提取性能在自建检测系统中存在一定差异,实验室在自建核酸检测系统时,选择核酸提取试剂盒时应进行充分验证,以保证检测结果的准确性。
Objective To compare the performance differences of different brands of nucleic acid extraction kits in self-built SARS-CoV-2 nucleic acid detection,and to provide reference for clinical laboratories to build their own SARS-CoV-2 nucleic acid detection systems.Methods The third-party SARS-CoV-2 nucleic acid detection quality control material was diluted to 4 concentrations.Nucleic acid extraction reagents from 4 different manufacturers were used:Shanghai Zhijiang Biotechnology Co.,Ltd.(A),Chongqing Zhongyuan Biotechnology Co.,Ltd.(B),Guangzhou Hexin Biotechnology Co.,Ltd.(C),and Hangzhou Bioer Technology Co.,Ltd.(D).It consisted of 4 self-built detection systems for detection.The positive detection rate and intra-assay repeatability of 4 self-built detection systems were compared.Results Among the samples of different concentrations,the samples extracted by the D brand extraction kit had the highest positive rate and the highest target gene detection rate.The Ct value of the amplification result of B brand was the smallest,and the concentration of the extracted product was the highest.Conclusion There are certain differences in the extraction performance of different brands of nucleic acid extraction reagents in the self-built detection systems.When laboratories build their own nucleic acid detection systems,the selection of nucleic acid extraction kits should be fully verified,to ensure the accuracy of the detection results.
