摘要
目的研究结肠癌组织中脑源性神经营养因子(NENF)的表达意义。方法利用蛋白质组学和公共数据库探索结肠癌蛋白质表达的变化,借助组织芯片进一步验证NENF在结肠癌中的表达及预后意义。结果蛋白质组学分析结肠癌与癌旁组织共得到429种差异表达蛋白,其中333种蛋白质上调,96种蛋白质下调。结肠癌中上调蛋白主要富集的通路有PI3K-AKT、磷脂酰肌醇信号系统、磷脂酶D、mTOR、MAPK等。NENF在结肠癌中高表达,且在周围神经侵犯阳性组中的表达量较阴性组高。组织芯片结果显示,NENF在结肠癌组织中高表达,且在Ⅱ、Ⅲ期样本中表达更高,较高的NENF表达与患者不良预后相关。结论在结肠癌中,NENF异常高表达,且与肿瘤分期和不良预后显著相关。
Objective To study the expression significance of neuron-derived neurotrophic factor(NENF)in colon cancer.Methods Proteomics and public databases were used to explore the changes of protein expression in colon cancer,and tissue microarray was used to further verify the expression and prognostic significance of NENF in colon cancer.Results Proteomic analysis of colon cancer and adjacent tissues yielded a total of 429 differentially expressed proteins,of which 333 were up-regulated and 96 were down-regulated.The main enriched pathways of up-regulated proteins in colon cancer were PI3K-AKT,phosphatidylinositol signaling system,phospholipase D,mTOR,MAPK,and so on.NENF was highly expressed in colon cancer,and the expression of NENF in the positive group of peripheral nerve invasion was higher than that in the negative group.The tissue microarray results showed that NENF was highly expressed in colon cancer tissues,and was even higher in stage Ⅱ and Ⅲ samples.Higher NENF expression was associated with poor prognosis of patients.Conclusion In colon cancer,NENF is abnormally highly expressed and significantly correlated with tumor stage and poor prognosis.
