摘要
建立了一种简便、快速同时测定乳清粉和乳粉中N-乙酰神经氨酸(Neu5Ac)和N-羟乙酰神经氨酸(Neu5Gc)的离子色谱分析方法。利用单因素变量法对酸解条件进行优化,借助固相萃取柱去除杂质干扰和溶剂转换,采用外标法定量。试验结果表明:采用0.05mol/L的硫酸溶液80℃水解1h,Dionex CarboPac PA20作为色谱柱洗脱,效果较为理想。N-乙酰神经氨酸(Neu5Ac)和N-羟乙酰神经氨酸(Neu5Gc)分别在0.03μg/mL~3μg/mL、0.00208μg/mL~0.208μg/mL内线性关系良好,样品平均回收率在96.6%~101.2%之间,重复性试验RSD值均小于5%。本方法简便、分析速度快、检测灵敏度高、重复性好、结果准确可靠,适用于乳清蛋白粉和奶粉中N-乙酰神经氨酸(Neu5Ac)和N-羟乙酰神经氨酸(Neu5Gc)的含量测定。
A simple and rapid ion chromatographic analysis method of N-acetylneurine(Neu5Ac)and Neu5Gc)in whey powder was established.Single factor variable method was used to optimize the acidolysis conditions,and impurity interference and solvent conversion were removed by solid phase extraction column.External standard method was used for quantification.The experimental results showed that the Dionex CarboPac PA20 column elution with 0.05mol/L sulfuric acid solution hydrolyzed at 80℃for 1h had a better effect.The linear ranges of Nacetylneuraminic acid(Neu5Ac)and N-hydroxylacetylneuraminic acid(Neu5Gc)were 0.03~3μg/mL and 0.00208-0.208μg/mL,respectively.The average recoveries were between 96.6%and 101.2%.RSDs of reproducibility test were less than 5%.This method is simple,rapid,sensitive,repeatable,accurate and reliable.It is suitable for the content determination of N-cetylneuraminic acid(Neu5Ac)and N-hydroxylacetylneuraminic acid(Neu5GC)in whey protein powder and milk.
