摘要
明确3种含笑属植物叶绿体基因组密码子的使用偏性及影响其偏性的因素。利用CodonW1.4.2、Genious 8.1.3、CUSP等软件对3个含笑属植物叶绿体基因组密码子的使用模式偏好性进行了分析。3种含笑属植物叶绿体基因组的GC含量在38.9%~39.3%之间;ENC值为在36.22~57.91之间,表明其密码子偏好性较弱。中性绘图、ENC-plot、PR2-plot分析表明影响3种含笑属植物叶绿体基因组密码子使用偏性的主要因素是自然选择。在此基础上,利用RSCU和ENC值筛选出3种含笑属植物叶绿体基因组的最优密码子共30个,其中29个以A/U结尾,UAU、CAU、CAA、AAU、AAA、GGA为3种含笑属植物共有的最优密码子。3种含笑属植物密码子偏好使用A/T结尾,其密码子使用偏性主要受到自然选择的影响。本研究对3种含笑属植物叶绿体基因组密码子的使用偏性进行分析,揭示影响其密码子使用偏性的主要因素,为后续3种含笑属叶绿体基因组外源蛋白表达载体的构建及优化等方面的研究提供了理论依据。
To clarify the usage bias of the chloroplast genome and the factors influencing the codon bias of three species of Michelia spp..In this study,the usage pattern preference of chloroplast genome codons of three species of Michelia was analyzed by CodonW1.4.2,Genious 8.1.3,CUSP and other software.The content of GC in the chloroplast genome of the three species of Michelia ranged from 38.9%to 39.3%.The ENC value ranged from 36.22 to 57.91.These indicate that the codon bias was weak.The neutral plot,ENc-plot and PR2-plot analysis showed that natural selection was the main factor affecting the codon usage bias of the chloroplast genome of the three species of Michelia spp..On this basis,30 optimal codons of chloroplast genomes of the three species of Michelia spp.were screened by RSCU and ENC values,among which 29 codons ended in A/U.UAU,CAU,CAA,AAU,AAA and GGA were the optimal codons of the three species of Michelia spp..The codon usage bias of the three species of Michelia spp.,which prefer to use the A/T ending,was mainly influenced by natural selection.In this study,by analyzing the bias in the use of chloroplast genomic codons in three species of the genus Michelia spp.,It lays a theoretical basis for the construction and optimization of heterologous protein expression vectors of chloroplast genomes in three species of Michelia spp..
