摘要
目的探索长链非编码RNA CCAT1对子宫内膜癌HEC-1A细胞增殖和侵袭的影响及可能机制。方法体外培养子宫内膜癌HEC-1A细胞,将lncRNA CCAT1抑制物转染子宫内膜癌HEC-1A细胞,抑制CCAT1表达,实时定量PCR检测各组细胞CCAT1和miR-218的表达,双荧光素酶报告基因实验分析CCAT1和miR-218的靶向关系。CCK-8法检测HEC-1A细胞增殖能力,Transwell实验检测HEC-1A细胞侵袭能力。结果转染CCAT1抑制物能够显著降低HEC-1A细胞中CCAT1的表达,上调miR-218的表达(P<0.05);双荧光素酶实验表明CCAT1可直接靶向miR-218。转染CCAT1抑制物后,HEC-1A细胞增殖和侵袭能力下降(P<0.05)。结论抑制lncRNA CCAT1可以使子宫内膜癌细胞HEC-1A的增殖和侵袭能力下降,其机制可能与上调miR-218的表达有关。
Objective To investigate the effect and mechanism of long-chain non coding RNA CCAT1 on the proliferation and invasion of endometrial carcinoma HEC-1 A cells.Methods The endometrial carcinoma HEC-1 A cells were cultured,and the CCAT1 inhibitor was transfected into HEC-1 A cells,the expression of lncRNA CCAT1 and miR-218 was detected by quantitative real-time PCR,and the target relationship between lncRNA CCAT1 and miR-218 was verified by double luciferase assay.The proliferation ability of HEC-1 A cells was detected by CCK-8,and the invasion ability of HEC-1 A cells was detected by Transwell assay.Results The transfection of CCAT1 inhibitor could significantly reduce the expression of CCAT1 and up-regulate the expression of miR-218(P<0.05).The dual luciferase assay showed that CCAT1 could directly target miR-218.After transfection of CCAT1 inhibitor,the proliferation and invasion ability of HEC-1 A cells was decreased(P<0.05).Conclusion Inhibition of lncRNA CCAT1 can reduce the proliferation and invasion of endometrial cancer cell HEC-1 A,and its mechanism may be related to up-regulate the miR-218.
