摘要
以发育成熟的掌状海带(Laminaria digitata)孢子体为材料,进行游孢子的放散、附着,形成胚孢子后,分别在不同的温度、光强和营养盐(氮、磷)浓度条件下培养,观察配子体生长发育和有性生殖的过程和特征,探究其配子体形成、排卵以及幼孢子体形成的最适环境条件。结果显示:氮浓度为4 mg/L、磷浓度为0.4 mg/L的营养盐浓度有利于掌状海带配子体的形成和成熟及幼孢子体的形成,营养盐浓度较低(氮≤1 mg/L,磷≤0.1 mg/L)或较高(氮=8 mg/L,磷=0.8 mg/L)均显著降低掌状海带的配子体形成率、排卵率以及幼孢子体形成率;15℃组的配子体形成率显著高于其它温度组,排卵率稍低于10℃组,15℃、光强≤40μmol photons/(m^(2)·s)组的幼孢子体形成率显著低于10℃组;20℃下,无排卵及幼孢子体形成;温度15℃、光强20μmol photons/(m^(2)·s)的条件下配子体形成率较高,而温度10℃、光强40μmol photons/(m^(2)·s)的条件下配子体排卵率和幼孢子体形成率显著较高。上述结果表明,掌状海带生殖过程中不同阶段对温度和光强的需求有较大差异。本研究获得了掌状海带有性生殖不同阶段适宜的温度、光强和营养盐(氮、磷)条件,可为掌状海带人工苗种繁育和养殖提供理论依据。
Using mature sporophyte of Laminaria digitata as material,the zoospores are released and attached to form embryo spores,and then cultured under different environmental factors.To explore the optimal environmental conditions for gametophyte formation,ovulation and young sporophyte formation of L.digitata,the process and characteristics from embryo spore to seedling formation were observed.The results showed:nitrogen concentration of 4 mg/L,phosphorus concentration of 0.4 mg/L,temperature of 15℃,light intensity of 20μmol Photons/(m^(2)·s)was the optimal conditions for gametophyte formation;Nitrogen:phosphorus concentration of 4 mg/L:0.4 mg/L,temperature of 10℃,light intensity of 40μmol Photons/(m^(2)·s)were the optimal condition for L.digitata gametophyte ovulation;Under the nitrogen concentration of 4 mg/L,phosphorus concentration of 0.4 mg/L,temperature of 10-15℃,light intensity of 40μmol Photons/(m^(2)·s),the formation of L.digitata young sporophyte was most favorable.This study indicates that the optimal environmental conditions at different growth and development stages of L.digitata are different.The data obtained in this study can provide theoretical basis and data support for artificial seed breeding and breeding of L.digitata.
