电针百会、神庭、足三里对胫骨骨折小鼠术后学习记忆能力的影响 被引量:1

摘要 目的探讨穴位电刺激对胫骨骨折内固定术后小鼠学习记忆能力、海马内α7nAChR表达及大脑炎症反应的影响。方法75只6月龄C57BL/6J雄性小鼠随机分为5组,每组各15只:对照组、胫骨骨折组(模型组)、穴位电刺激+胫骨骨折组(电针组)、α7nAChR激动剂PNU282987+胫骨骨折组(PNU组)及穴位电刺激+α7nAChR拮抗剂MLA+胫骨骨折组(MLA+电针组)。75只小鼠参与水迷宫定位巡航训练1周后对模型组、电针组、PNU组和MLA+电针组小鼠做胫骨骨折内固定模型处理。造模后第1天开始对各组小鼠加用相应刺激条件,直至处死当天。分别于术前1 d,术后1,3,7 d分批处死小鼠,术前1 d,术后1,3 d每组各处死3只,术后7 d每组各处死6只。整个实验采用定位巡航实验和空间探索试验评估小鼠学习记忆能力,酶联免疫吸附试验法检测小鼠海马内IL-4、IL-10、IL-1β、TNF-α含量,免疫荧光实验检测小鼠海马内α7nAChR表达情况。结果定位航行实验显示模型组在术后1,3,7 d逃避潜伏期明显大于对照组(P<0.05),表明小鼠胫骨骨折内固定模型建立成功;术后1,3,7 d电针组逃避潜伏时间明显小于模型组(P<0.05)。空间探索实验显示术后7 d电针组穿越平台次数明显多于模型组(P<0.05)。ELISA实验结果显示电针组在术后1,3,7 d的IL-1β、TNF-α含量明显小于模型组(P<0.05);电针组术后1,3,7 d的IL-4、IL-10含量明显多于模型组(P<0.05)。免疫荧光实验结果显示电针组海马内α7nAChR含量明显多于模型组(P<0.05)。结论穴位电刺激可以增加海马内α7nAChR含量,减弱大脑炎症反应并且改善小鼠胫骨骨折术后的学习记忆能力。 Objective To investigate the effects of acupoint electrical stimulation on learning and memory ability,α7 nAChR expression and brain inflammatory response in hippocampus of mice after internal fixation of tibial fracture.Methods Seventy-five 6-month-old male C57 BL/6 J mice were randomly divided into five groups:control group,tibial fracture model group(model group),acupoint electrical stimulation+tibial fracture group(EA group),α7 nAChR agonist PNU282987+tibial fracture group(PNU group)and acupoint electrical stimulation+α7 nAChR antagonist MLA+tibial fracture group(MLA+EA group).After seventy-five mice participated in water maze positioning cruise training for one week,the tibial fracture internal fixation models were established in model group,EA group,PNU group and MLA+EA group.On the first day after modeling,the mice in each group were given the corresponding stimulation conditions until the day of death.Three mice in each group were executed at day 1 before operation,at day 1,3 after operation,and six mice in each group were executed at 7 d after operation.In the whole experiment,the learning and memory ability of mice was evaluated by place navigation test and spatial probe test,and IL-4,IL-10 and IL-1β,TNF-αin hippocampus were detected by enzyme-linked immunosorbent assay,and the expression ofα7 nAChR in hippocampus was detected by immunofluorescence assay.Results The escape latency in model group was significantly higher than that in control group at day 1,3,7 after ope-ration(P<0.05),indicating that the internal fixation model of mouse tibial fracture was successfully established.The escape latency in EA group was significantly shorter than that in model group at day 1,3,7 after operation(P<0.05).The number of crossing the platform in EA group was significantly higher than that in model group at day 7 after operation(P<0.05).ELISA results showed that the contents of IL-1β,TNF-αin EA group were significantly lower than that in model group at day 1,3,7 after operation(P<0.05),while the contents of
